Granulosa cells play important roles in the regulation of ovarian functions. Phospholipase C is crucial in several signalling pathways and could participate in the molecular mechanisms of cell proliferation, differentiation and ageing. The objective of this study was to identify the effects of phospholipase C on the steroidogenesis of oestradiol and progesterone in porcine granulosa cells cultured in vitro. Inhibitor U73122 or activator m‐3M3FBS of phospholipase C was added to the in vitro medium of porcine granulosa cells, respectively. The secretion of oestradiol decreased after 2 hr, 8 hr, 12 hr, 24 hr and 48 hr of treatment with 500 nM U73122 (p < .05) and decreased after 2 hr of treatment in the 500 nM m‐3M3FBS addition group (p < .05). The secretion of progesterone increased after 4 hr of treatment with 500 nM U73122 (p < .05) and increased after 2 hr and 8 hr of treatment in the 500 nM m‐3M3FBS addition group (p < .05). The ratio of oestradiol to progesterone decreased at each time point, except 8 hr after the addition of 500 nM U73122 (p < .05). The ratio of oestradiol to progesterone decreased after 2 hr (p < .05) of treatment with 500 nM m‐3M3FBS. In genes that regulate the synthesis of oestradiol or progesterone, the mRNA expression of CYP11A1 was markedly increased (p < .05), and the mRNA expression of other genes did not change significantly in the U73122 treatment group, while the addition of m‐3M3FBS did not change those genes significantly despite the contrary trend. Our results demonstrated that phospholipase C can be a potential target to stimulate the secretion of oestradiol and suppress progesterone secretion in porcine granulosa cells cultured in vitro, which shed light on a novel biological function of phospholipase C in porcine granulosa cells.

Zearalenone (ZEN) is a mycotoxin that not only binds to estrogen receptors, but also interacts with steroidogenic enzymes and acts as an endocrine disruptor. The aim of this study was to verify the hypothesis that low doses, minimal anticipated biological effect level (MABEL), no-observed-adverse-effect level (NOAEL) and lowest-adverse-effect level (LOAEL), of ZEN administered orally for 42 days can induce changes in the peripheral blood concentrations of selected steroid hormones (estradiol, progesterone and testosterone) in pre-pubertal gilts. The experiment was performed on 60 clinically healthy gilts with average BW of 14.5 ± 2 kg, divided into three experimental groups and a control group. Group ZEN5 animals were orally administered ZEN at 5 μg ZEN/kg BW, group ZEN10 — at 10 μg ZEN/kg BW, group ZEN15 — at 15 μg ZEN/kg BW, whereas group C received a placebo. Five gilts from every group were euthanized on analytical dates 1, 2 and 3 (days 7, 14 and 42 of the experiment). Qualitative and quantitative changes in the biotransformation of low ZEN doses were observed. These processes were least pronounced in group ZEN5 (MABEL dose) where ZEN metabolites were not detected on the first analytical date, and where β-ZEL was the predominant metabolite on successive dates. The above was accompanied by an increase in the concentration of estradiol (E2) which, together with “free ZEN”, probably suppressed progesterone (P4) and testosterone (T) levels.

Background: The hormonal control of ovulation has become a standard procedure in the swine industry. However, exogenous gonadotropins can be detrimental to reproductive function, affecting follicle development, corpus luteum formation, and embryo development and survival. Much less is known about uterine receptivity in gilts with induced estrus. Therefore, our objective was to determine the effect of estrus induction with pregnant mare serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG) on the expression of steroid, prostaglandin, cytokine, and oxytocin receptors, as well as nuclear factor kappa B subunit 1 (NFKB1), peroxisome proliferator activated receptor gamma (PPARG), and gap junction protein alpha 1 (GJA1), in the endometrium and myometrium of early pregnant gilts. Twenty prepubertal gilts received 750 IU PMSG and 500 IU hCG 72 h later, while eighteen prepubertal gilts in the control group were observed daily for estrus behavior. All gilts were inseminated in their first estrus and slaughtered on days 10, 12, and 15 of pregnancy to collect uterine tissues for mRNA expression analyses using real-time PCR.Results: Estrus induction did not affect progesterone receptor expression in either uterine tissue. In the endometrium, greater mRNA expression of estrogen receptors (ESR1 and ESR2), androgen receptor (AR), prostaglandin (PG) E2 receptors (PTGER2 and PTGER4), PGF2α receptor (PTGFR), interleukin 6 receptor (IL6R), tumor necrosis factor α receptors (TNFRSF1A and TNFRSF1B), and oxytocin receptor (OXTR) was detected in the control than in the PMSG/hCG-treated gilts (P < 0.05). In the myometrium, concentrations of AR, PTGER2, PTGFR, and NFKB1 transcripts were lower, while PGI2 receptor and PPARG transcripts were elevated in gilts with gonadotropin-induced estrus as compared with naturally ovulated gilts (P < 0.05). Furthermore, the administration of PMSG/hCG resulted in the greater expression of GJA1 mRNA in both the endometrium and myometrium of day 15 pregnant gilts (P < 0.05). Conclusions: Estrus induction with PMSG/hCG in prepubertal gilts may affect steroid, prostaglandin, cytokine, and oxytocin receptor expression in the endometrium and myometrium, thereby altering uterine receptivity to local or systemic factors. This may, in turn, contribute to disorders in embryo-maternal interactions and the process of implantation.
