Role of Caspase-1 in the Pathogenesis of Inflammatory-Associated Chronic Noncommunicable Diseases

Abstract: Caspase-1 is the first and extensively studied inflammatory caspase that is activated through inflammasome assembly. Inflammasome is a cytosolic formation of multiprotein complex that aimed to start inflammatory response against infections or cellular damages. The process leads to an auto-activation of caspase-1 and consequent maturation of caspase-1 target molecules such as interleukin (IL)-1β and IL-18. Recently, the role of caspase-1 and inflammasome in inflammatory-induced noncommunicable diseases (NCDs) l… Show more

“…Molecular docking analysis was performed to provide further insight into the molecular mechanism of the anti-inflammatory capacity of TA against rotenone-induced inflammation. Tumour necrosis factor-alpha (TNF-α), Cyclooxygenase-2 (Cox-2) and Caspase-1 (Casp-1) are the pro-inflammatory mediators accompanied with NO release, through NF-kB and inflammasome complex-mediated responses 43 , 44 . These proteins are members of the group of pro-inflammatory markers that might be stimulated or released by glial cells, as a result of the signalling interplays of their molecular origins, during chronic neuroinflammation-induced PD 45 47 .…”

“…NF- κ B primes the NLRP3-inflammasome for activation by inducing pro-IL-1 β and NLRP3 expression 33 . On the one hand, the NF- κ B signaling pathway activates and upregulates the transcription and translation of pro-IL-1 β ; on the other hand, the NLRP3 inflammasome promotes the maturation of cleaved-caspase-1, and cleaved pro-IL-1 β activates cleaved-IL-1 β 10 , 34 . The activated cleaved-IL-1 β , as an indispensable upstream inflammatory factor, mediates the upregulation of a variety of inflammatory factors, mediating the cascade inflammatory response of lung tissue cells 35 .…”

“… 39 - 41 The expression of IL-1β, MMP13, and caspase-1 is elevated in the OA cartilage. 42 - 45 Varani et al. 46 and Ongaro et al 47 reported that at 1.5 mT, the 75-Hz PEMF increased anti-inflammatory properties in chondrocytes via mediating an upregulation of A2A and A3 receptors and inhibiting the release of PGE2, IL-1β, and IL-6 production.…”
