lncRNA NEAT1 Facilitates Cell Proliferation, Invasion and Migration by Regulating CBX7 and RTCB in Breast Cancer

Abstract: To investigate the association between the lncRNA NEAT1 and breast cancer, and to determine the influence of NEAT1 on regulation of other signaling molecules in breast cancer. Methods: In the present study, we measured levels of the lncRNA NEAT1 in 106 breast cancer patients and in a human breast cancer cell line by qRT-PCR. The correlation between NEAT1 expression and patients' clinical characteristics was analyzed with in-house and TCGA data. We used cellular functioning assays and cell immunofluorescence as… Show more

“…We suggest a possible reciprocal relationship between MALAT1 and each of AC005154.1, NEAT1, AL139022.1, AC007622.2, USP3-AS1, MUC20-OT1, LINC-PINT, and AC103810.5. In fact, several studies on NEAT1 lncRNA have shown that upregulated NEAT1 facilitated tumor cell viability, invasion, and migration in breast cancer 32 . In Non-small-cell lung carcinoma, Shikonin mediated NEAT1 suppression resulted in apoptotic cell death of paclitaxel-resistant cell lines 33 .…”

“…21 lncRNA NEAT1 has been reported to regulate the growth and mobility of BC cells by targeting CBX7 and RTCB. 22 Also, lncRNA NEAT1 confers chemoresistance and cell "stemness" in triple-negative breast cancer (TNBC) cells. 23 Epithelial-mesenchymal transition (EMT) is a key inducer of tumor metastasis, 24 but the effect of lncRNA NEAT1 on EMT in BC cells is not known.…”

“…Breast Cancer Bioinformatic analysis of Chromobox proteins family has shown that the most significant difference between breast cancer and normal mammary tissue is the low mRNA expression of CBX7 (Li X. et al, 2020). One study of breast cancer implied that patients with low CBX7 had lower survival and were more likely to develop lymph node metastasis, P53 mutations, and the cancer further deteriorated, metastasized over time (Li X. et al, 2020). The mechanism for its carcinogenicity may be that CBX7 acts as a novel epigenetic regulator of the Wnt/β-catenin pathway in breast cancer to determine cancer progression (Li X. et al, 2020).…”

The mechanism for its carcinogenicity may be that CBX7 acts as a novel epigenetic regulator of the Wnt/β-catenin pathway in breast cancer to determine cancer progression (Li X. et al, 2020). In breast cancer, CBX7 increases Dickkopf-1 (DKK-1, a Wnt antagonist) gene transcription, thus indirectly affecting the Wnt signaling pathway (Bafico et al, 2001;Kim et al, 2015).…”
