Edaravone Ameliorates Renal Warm Ischemia-Reperfusion Injury by Downregulating Endoplasmic Reticulum Stress in a Rat Resuscitation Model

Abstract: Background: This study was conducted to explore whether the effect of edaravone (5-methyl-2-phenyl-2,4-dihydro-3H-pyrazol3-one, EDR) can ameliorate renal warm ischemia-reperfusion injury (IRI) by modulating endoplasmic reticulum stress (ERS) and its downstream effector after cardiac arrest (CA) and cardiopulmonary resuscitation (CPR) in a rat model. Methods: The rats (n=10) experienced anaesthesia and intubation followed by no CA inducement were defined as the Sham group. Transoesophageal alternating current s… Show more

“…6−8 Antioxidants could ameliorate AKI through inhibition of oxidative stress. 9 Curcumin (Cur) is a natural diphenolic compound with specific antioxidant and antiinflammatory properties. Cur exerted a reno-protective effect in AKI via relieving intracellular superoxide generation, increased superoxide dismutase levels, inhibition of oxidative stress cellular apoptosis, and inducing autophagy.…”

“…Aberrant and excessive reactive oxygen species (ROS) in AKI lead to oxidative stress, inflammation, and vascular damage. Antioxidants could ameliorate AKI through inhibition of oxidative stress . “…Although appropriate ROS is essential for intracellular signaling and immune response against pathogens, the excessive ROS can not be entirely scavenged by cellular antioxidants, which would mediate the severe damage on cells by destroying organelle, cell membrane, and nuclei DNA (Nita and Grzybowski, 2016 ). To protect injured tissues and prevent further damage of organs during transplantation, the administration of antioxidant reagents for scavenging ROS has been widely applied, such as edaravone (Fu et al, 2020 ). However, the usage of edaravone is generally accompanied by a series of side effects ranging from bruising to skin inflammation or rash, leading to many concerns and demands of new types of antioxidant reagents.…”

“…Specifically, the use of vanillin has been proposed to reduce kidney damage and cisplatin-induced nephrotoxicity [30]. Similarly, the use of EDA is also envisioned to prevent cisplatin-induced acute kidney injury [31] and as a reno-protective drug in diverse pathological situations [32][33][34]. Both EDA and vanillin can be useful protecting agents to reduce and prevent kidney diseases, but our work strongly suggests that the two compounds shall never be combined and used simultaneously, due to the high risk of adducts formation.…”
