In Vitro Analysis of the Immunomodulating Effects of Allium Hookeri on Lymphocytes, Macrophages, and Tumour Cells

Abstract: The present study investigated the effects of ethanol extracts of Allium hookeri (leaf, root, and fermented root) on parameters of innate immunity, tumour cell viability and antioxidant effect in vitro. Innate immunity was measured by spleen lymphocyte proliferation, nitric oxide production by chicken macrophage HD11 cells and suppressive effect on tumour cell viability was assessed using chicken RP9 cells. Free radical scavenging capacity as a measure of antioxidant capacity was determined by 0.15 mM of DPPH … Show more

“…Total phenolic content (TPC) analysis was modified from the method of Senhaji et al [ 13 ]. Twenty microliters of samples (50, 100, 250, 500, and 1000 μg/mL) were mixed with 80 μL of distilled water and 40 μL of 100% Folin–Ciocalteu’s phenol reagent (Sigma-Aldrich Co., St. Louis, MO, USA) in a 96-well plate, and it was reacted for 3 min.…”

“…Organosulfur compounds are considered major chemical markers in AH, showing anti-inflammatory and anti-obesity in previous studies [ 9 , 10 , 11 ]. However, there is only limited information available on the anti-oxidant and immune stimulatory effects of AH leaves [ 13 , 14 ] compared with those of AH roots.…”

“…As these components exhibit antioxidant activities, the use of A. hookeri is promising as a medical food [14,15]. A. hookeri exhibits immunomodulatory effects in lymphocytes, macrophages, and tumor cells in in-vitro chicken cell experiments [16]. In vivo experiments have also suggested that A. hookeri inhibits the inflammatory response in the pancreas of diabetic rats and LPS-induced young broiler chickens [17,18].…”

“…The spleen lymphocyte transformation capacity can reflect the state of the body's cellular immune function and malignant tumors lead to a decrease in lymphocyte transformation capacity (Lee et al., 2017). The spleen lymphocyte transformation capacity of all test groups was significantly enhanced in a dose‐dependent manner compared with the capacity of the TC group (Table 3).…”
