LPS- and LTA-Induced Expression of IL-6 and TNF-αin Neonatal and Adult Blood: Role of MAPKs and NF-κB

Abstract: As nuclear factor kappa B (NF-κB) and mitogen-activated protein kinases (MAPKs) seem to be critical mediators in the inflammatory response, we studied the effects of lipopolysaccharide (LPS) and lipoteichoic acid (LTA) on (a) the activation of NF-κB and MAPKs and (b) the expression of tumor necrosis factor alpha (TNF-α) and interleukin 6 (IL-6) with or without the specific inhibitors of these intracellular signal transduction pathways in neonatal cord and adult blood. TNF-α and IL-6 concentrations showed a sha… Show more

“…Previous studies have indicated that aLTA can induce the phosphorylation of p65, thereby activating the NF-B signaling pathway, upregulating the expression of TNF-␣, and subsequently inducing inflammatory responses in host cells [27,28]. We found that aLTA induced p65 mRNA expression, significantly enhanced p65 phosphorylation, and upregulated the expression of TNF-␣ in HT-29 cells.…”

“…When activated by external stimuli, p38, JNK, and ERK phosphorylation is increased and related proinflammatory cytokines will begin to be expressed [30]. Furthermore, the present study explores the anti-inflammatory mechanism of Andrographolide regarding the NF- κ B and MAPKs pathways.…”
