Low-molecular-weight RNAs and initiation of RNA-directed DNA synthesis in avian reticuloendotheliosis virus

Abstract: The small RNAs of avian reticuloendotheliosis virus (REV) were analyzed by two-dimensional polyacrylamide gel electrophoresis and compared with those of murine leukemia virus and avian sarcoma virus. Although there were some similarities among the three virus types, the patterns of small RNAs were distinct. By characterizing the small RNA which is most tightly associated with REV genome RNA and which can be labeled in limited DNA synthesis reactions, the primer for REV reverse transcription was identified as t… Show more

“…In every case, the products obtained conformed to the proposed MuMTV particles were found to contain a subset of the host cell tRNA population. However, this subset was restricted to only two major species rather than the 10 or more different tRNA's observed in other viral systems (28,29,34). Both of these tRNA's were specific for the amino acid lysine, confirming the previous findings of Waters (41).…”

“…After incubation at 37°C for 1 h, the reaction products were diluted in buffer containing 10 mM Tris-hydrochloride (pH 7.6), 100 mM NaCl, 10 mM EDTA and 0.2% sodium dodecyl sulfate, phenol extracted once, and ethanol precipitated. The recovered products were analyzed by electrophoresis in one dimension in 0.3-mm slab gels of 8% acrylamide containing 7 M urea (28). Any [a-32P]dNMP-tagged products were eluted from the gel and further characterized by 2-D gel electropho-RESULTS Low-molecular-weight RNAs of MuMTV.…”

“…REV-A REV-T c-re/ (turkey) primer associated with virion RNA of reticuloendotheliosis viruses and murine leukemia viruses (15,33). In contrast, avian leukosis-sarcoma viruses have tRNATTrp primers (7).…”

“…The taxonomical position of the REV group within the Retroviridae is uncertain (10,51). However, there is evidence that the REVs are related to mammalian retroviruses, all of which use tRNA Pro to initiate reverse transcription (38). The capsid and reverse transcriptase proteins share antigenic similarity (1, 3-5, 8, 18, 36).…”
