The biological characteristics of three‐dimensional (3D) tumors cannot be adequately represented through two‐dimensional (2D) cell cultures. Here, to acquire more precise information about plasma–tumor interactions, the plasma‐induced cytotoxicity was tested in both 2D cell culture and 3D tumor‐spheroid culture. The results indicated that the toxic effects were markedly reduced in 3D cancer cells compared with 2D cells. We found that reactive oxygen and nitrogen species can penetrate through the entire depth of 3D tumor spheroid, but local apoptosis was observed only on the outermost layer. The permeation depth of plasma‐generated reactive species was then tracked in 3D tumor spheroids. We hypothesized that plasma‐induced apoptosis within a tumor depends on the “deep” delivery of H2O2, NO2−, and NO3− rather than O2−, OH•, and ONOOH/ONOO−.

Cold atmospheric plasma (CAP) is a novel technology that generates a unique combination of reactive oxygen and nitrogen species, electric fields, and UV radiation. CAP has shown promise in regulating the immune system and has potential clinical applications in wound healing, cancer treatment, and infection control. This review provides an overview of the immunological regulation activity of CAP, highlighting its substantial impact on cytokines production, immune cell phagocytosis, and immune cell proliferation. CAP has also been demonstrated to have potent therapeutic effect in anti‐inflammation, wound repair, viral and bacterial infections. Furthermore, CAP has been investigated as an adjuvant therapy for tumor treatment, eliciting a robust antitumor immune response and remarkable synergistic effects in diverse combination therapies. Further research is needed to fully understand the mechanisms underlying the effects of CAP on the immune system and to optimize its clinical application.This article is protected by copyright. Cold plasmas are continuously developed for biomaterials engineering as well as for therapeutic treatments of cells and tissues. For this last application plasma activated media (PAM) and reactive oxygen and nitrogen species (RONS) gained attention as key players. Here, the use of cold atmospheric pressure (AP) plasma is described, for generating RONS in Dulbecco's Modified Eagles Medium (DMEM); superoxide anion (normalO2−), hydrogen peroxide (H2O2), nitrates (NO3−), and nitrites (NO2−) were detected. PAM was applied to Bone Marrow Stem Cells (BMSC) and SAOS‐2 osteoblasts. Both native and plasma‐modified polymeric scaffolds were used as three dimensional (3D) supports for cell cultures. The cell activity was found dependent on both PAM and cell type. BMSCs grown on plasma‐coated scaffolds tolerated better PAM with respect to those on native scaffolds.
