Loss of the Drosophila melanogaster DEAD box protein Ddx1 leads to reduced size and aberrant gametogenesis

Abstract: Mammalian DDX1 has been implicated in RNA trafficking, DNA double-strand break repair and RNA processing; however, little is known about its role during animal development. Here, we report phenotypes associated with a null Ddx1 (Ddx1(AX)) mutation generated in Drosophila melanogaster. Ddx1 null flies are viable but significantly smaller than control and Ddx1 heterozygous flies. Female Ddx1 null flies have reduced fertility with egg chambers undergoing autophagy, whereas males are sterile due to disrupted sperm… Show more

“…18 Because DDX1 deficiency in mice causes early embryonic lethality, it must play essential roles in normal cells. 19 DDX1 plays a critical role in testicular tumorigenesis in part by promoting transcription of Cyclin-D2 and stem cell-related genes on human chromosome 12p. 20 The expression level of DDX1 is elevated not only in germ cell tumors but also in retinoblastoma, neuroblastoma, glioblastoma and breast cancer.…”

“…DDX1 is involved in a variety of biological processes, including tRNA synthesis, mRNA and microRNA processing, ribosome biogenesis, DNA repair, and nuclear factor‐kappaB‐mediated gene induction . “…The DEAD box 1 gene (DDX1) is a widely expressed gene that is misexpressed in a number of cancers, including retinoblastoma, neuroblastoma, and breast cancer (1,2). Knockout of DDX1 leads to early embryonic lethality in mice and severely reduced fertility in flies (3,4). DDX1 is involved in the transport of RNAs from the nucleus to the cytoplasm and regulates cytoplasmic localization of the splicing-regulatory protein KSRP (5).…”

“…DDX1 null flies were infertile, significantly smaller, and developmentally delayed, with both oogenesis and spermatogenesis affected. Significantly smaller ovaries containing few to no mature eggs were observed in DDX1 null flies ( Germain et al., 2015 ). DDX1 was highly expressed in the developing oocytes and the surrounding granulosa cells of the ovaries.…”
