Loop Mediated Isothermal Amplification (LAMP) Test- a Novel Nucleic Acid Based Assay for Disease Diagnosis

Abstract: Disease diagnosis is of crucial importance in making appropriate therapeutic decisions. Conventionally, diagnosis was based on isolation and identification of the causative agent. This was followed in timeline by the development of serological tests. Nowadays, molecular tests credited with high sensitivity like polymerase chain reaction and its variants are replacing the older diagnostic techniques. Major obstacle in adoption of PCR based technique is the need for thermocycler, which precludes its use in low r… Show more

“…The amplified products can be detected by indirect detection methods like turbidity and non-specific dyes [53]. Compared to conventional PCR, LAMP assays are highly sensitive, rapid, cost-effective, and can be easily implemented in resource-limited settings [55]. However, complicated primer design and the possibility of carry-over contamination still remain a concern for LAMP-based assays [50,56].…”

“…Different techniques like Restriction Fragment Length Polymerase (RFLP) assay (Kumar et al, 2011b), Lateral Flow assay (Arun et al, 2014), Loop mediated Isothermal Amplification (LAMP) test (Rekha et al, 2014;Rekha et al, 2015) have been tried. Combination of conventional and advanced molecular tests is preferred for confirmation of field outbreaks.…”

“…This test provides a rapid and sensitive diagnosis in a rural environment, where no laboratory equipment was available. LAMP can be performed where thermocyclers cannot be easily maintained/ procured (Rekha et al, 2014). The advantage of these tests is no need of agarose gel electrophoresis.…”

“…(such as a thermocycler) and post amplification processing and are still time-consuming to accomplish at chair-side [3]; these drawbacks led to the emergence of the alternative isothermal amplification methods which are easier and not requiring thermal cycling as these methods amplify nucleic acids to large copies under isothermal conditions [4]. Several isothermal amplification protocols have been generated over the last 10 years, one of which loop-mediated isothermal amplification (LAMP).…”
