Epstein‐Barr virus (EBV) is a prevalent γ‐herpesvirus associated with a variety of cancers, including epithelial nasopharyngeal carcinoma (NPC) and gastric carcinoma (GC). Long noncoding RNAs (lncRNAs) are emerging as powerful regulators that have demonstrated to play crucial roles in cancer development. In this approach, based on genome‐wide RNA sequencing, we examined the lncRNA expression profiles in four EBV genome‐infected and EBV‐negative 293 cells. A series of lncRNAs were found to be dysregulated in a comparative analysis. Then, eight typical lncRNAs were selected for validation of expression levels by real‐time quantitative polymerase chain reaction and were detected in EBV‐positive or EBV‐negative GC and NPC cells. The differential expression patterns of the eight lncRNAs for validation were fundamentally identical to those revealed in RNA sequencing. Particularly, the differential expression of these lncRNAs in GC and NPC cells indicated their possible roles in EBV infection and tumorigenesis. In addition, a predicted lncRNA‐microRNA‐messenger RNA network suggested their potential interactions. This study reveals the first lncRNAome related to EBV infection in the epithelial cells and provides novel clues for the study of viral role in epithelial cancers through the interaction between EBV and host lncRNAs.

Abstract. Long non-coding RNAs (lncRNAs) have been identified as critical regulators in tumorigenesis. In our present study, we measured the level of small nucleolar RNA host gene 5 (SNHG5) in bladder cancer (BC) tissues and cell lines, and the correlation of the level of SNHG5 with clinicopathological features and prognosis of BC patients was analyzed. Reverse transcription-quantitative polymerase chain reaction was performed to determine the level of SNHG5 in the BC tissues and cell lines. The Kaplan-Meier method was used to analyze the long-term survival outcomes. MTT and colony formation assays were applied to assess the influence of SNHG5 on cell proliferation ability. Flow cytometry was used to measure the function of SNHG5 on cell cycle and apoptosis rate. SNGH5 was found upregulated in BC tissues and cell lines and a high level of SNGH5 was correlated with a poor prognosis. Silencing SNHG5 inhibited the proliferation ability of BC cells and such a function was attributed to its influence on cells cycle and apoptosis. Our findings imply that SNHG5 was upregulated in BC tissues and played an important role in BC progression and may be a potential therapeutic target for BC patients.

LncRNAs (long noncoding RNAs) are noncoding transcripts that are more than 200 nt long and have been described as the largest subclass in the noncoding transcriptome in humans. Although studies of lncRNAs in cancer have been continuing for a long time, no much has been known about X chromosome‐linked lncRNAs. Here, by using RNA‐seq we report the identification of a new X chromosome‐linked lncRNA (lnc‐XLEC1) that is aberrantly downregulated during the development of endometrial carcinoma (EC). The overexpression of lnc‐XLEC1 reduces the migration and proliferation of EC cells. Flow cytometry analysis indicated that lnc‐XLEC1 overexpression resulted in a substantial accumulation of EC cells in the G1 phase. In addition, lnc‐XLEC1 had inhibitive effects that may result from its collaboration with MBP‐1 during the suppression of the c‐Myc expression and the negative regulating of the Cdk/Rb/E2F pathway. The anti‐tumor effects of lnc‐XLEC1 on EC progression suggest that lnc‐XLEC1 has some potential value in anti‐carcinoma therapies and deserves further investigation. Our study reported for the first time that the lnc‐XLEC1 might be related to the incidence and prognosis of EC. Moreover, we discovered that this process might be related to somatic X dosage compensation and skewed X chromosome inactivation (SXCI).
