Long-chain omega3 polyunsaturated fatty acids and cognition in older people: interaction with APOE genotype

Abstract: -Basic research and epidemiological studies suggest a protective effect of long-chain omega3 polyunsaturated fatty acids (LC n-3 PUFA) against age-related cognitive decline. However, most randomized controlled trials with LC n-3 PUFA supplements have yielded disappointing results on cognitive outcomes in older persons. One explanation for this discrepancy may be an inadequate targeting of potential beneficiaries of LC n-3 PUFA according to their Apolipoprotein E (APOE) genotype. The aim of this paper was to ex… Show more

“…It is widely known that fatty acid metabolism and composition can be altered during diseases, resulting in beneficial ( 4 , 5 ) or adverse events ( 6 , 7 ). In this context, cardiovascular (CV) health may be affected by MUFAs through effects on various markers associated with CHD ( 8 ), including serum lipid and lipoprotein profile ( 9 ), vascular function markers ( 10 ), and postprandial vascular function ( 11 ).…”

“…Third is genetic factors: certain genetic mutations might impact the efficacy of omega-3. For instance, individuals carrying the APOE4 allele may not respond as well to omega-3 as others [ 21 , 52 ]. In conclusion, while omega-3 may be beneficial for certain populations, it does not necessarily benefit everyone.…”
