Localization of the syndactyly type II (synpolydactyly) locus to 2q31 region and identification of tight linkage to HOXD8 intragenic marker

Abstract: Syndactyly type II (SynPolyDactyly; SPD) is an autosomal dominant condition with incomplete penetrance and variable expressivity. Sixty-two meioses from a kindred with 425 individuals were used to map the SPD locus to 2q31 region, approximately 1.7 cM (Lod score = 12.96) centromeric to HOXD8 intragenic marker. Other homeobox-containing genes in this region have previously been ordered as cen-DLX1/DLX2-EVX2-(5' --> HOXD13..HOXD8.HOXD1 --> 3')-tel')-tel. A single recombinant with HOXD8 excluded the most 3' end o… Show more

“…Particular genes have been located on the 5’ region of the A- and D- clusters of human chromosomes 7 and 2 respectively [41]. These genes appear to influence limb patterning and of particular interest is the Homeobox D gene (HOXD), in particular related to the loci at 2q31 [65]. …”

“…Synpolydactyly (SPD) locus has been mapped to the 2q31 region, approximately 1.2 cM centromeric to the IIOXD8 intragenic marker in one large Turkish kindred (Sarfarazi et al, 1995b). This assignment has subsequently been confirmed in a second Turkish (Abstract 11) and a Mexican kindred.…”

“…Highly polymorphic simple sequence repeats comprising six loci containing candidate genes (Table II) were identified either by literature review [Sarfarazi et al, 1995;Lin et al, 1996;Li et al, 1997] or by searching the physical map, radiation hybrid map, and linkage map databases accessible through the National Center for Biotechnology Information (http://www. ncbi.nlm.nih.gov/).…”
