Localisation of clozapine during experimental autoimmune encephalomyelitis and its impact on dopamine and its receptors

Abstract: Multiple sclerosis is a disease characterised by axonal demyelination in the central nervous system (CNS). The atypical antipsychotic drug clozapine attenuates experimental autoimmune encephalomyelitis (EAE), a mouse model used to study multiple sclerosis, but the precise mechanism is unknown and could include both peripheral and CNS–mediated effects. To better understand where clozapine exerts its protective effects, we investigated the tissue distribution and localisation of clozapine using matrix-assisted l… Show more

“…In accordance with our nding, the expression of D2R on brain-resident or in ltrating immune cells in EAE was already showed to be signi cantly increasing[65]. Moreover, in the work byRobichon et al (2021) no signi cant reduction in the immunocontent of D1R on macrophages and monocytes was reported[65]…”

“…Clozapine and risperidone were tested in patients with MS (CRISP, or Community ReIntegration for Socially isolated Patients, trial, ACTRN12616000178448), but the trial was halted early due to a higher incidence of adverse side effects during the titration stage (72), prompting investigations into the pharmacodynamics of these drugs. Robichon et al (73) investigated the biodistribution of clozapine in a mouse EAE model using matrix-assisted laser desorption ionization, or MALDI, imaging (73). Higher levels (fivefold) of clozapine in the serum were observed in EAE mice compared with control mice.…”

“…Flow cytometry has not been traditionally used to measure receptor expression, but advancements in this technique have resulted in it being capable of measuring membrane-bound receptor expression in single intact cells. In fact, a procedure for measuring D2 receptor expression in rodents was recently optimised and published by the consulted research group (Robichon et al, 2021). Additionally, this group generously offered to provide both the technical support and laboratory supplies required to conduct flow cytometry.…”
