Local anesthetic lidocaine delivery system: chitosan and hyaluronic acid-modified layer-by-layer lipid nanoparticles

Abstract: Context: Transdermal local anesthesia is one of the most applied strategies to avoid systemic adverse effects; there is an appealing need for a prolonged local anesthetic that would provide better bioavailability and longer pain relief with a single administration. Objective: Layer-by-layer (LBL) technique was used in this study to explore a nanosized drug delivery system for local anesthetic therapy. Materials and methods: LBL-coated lidocaine-loaded nanostructured lipid nanoparticles (LBL-LA/ NLCs) were prep… Show more

“…Hyaluronate chitosan multilayer liposomes (containing 10 alternating layers) with 528 nm particle size exhibited an enhanced transdermal delivery than uncoated unilamellar liposomes (~100 nm) (Jeon et al, 2015). Similarly, multilayered nanostructured lipid nanoparticles coated with hyaluronic acid and chitosan (181 nm) showed a two-fold larger flux across rat skin compared to uncoated nanoparticles (Zhang et al, 2016). In contrast, other authors support that multilamellar and small unilamellar liposomes possess similar ability to penetrate into the SC and viable skin (Lymberopoulos, 2017).…”

“…In vitro skin permeation behaviors of NLCs and LPNs were examined using the Franz diffusion method (Zhang et al., 2016 ). The skins from the abdominal surface of rats (fur, adherent fat, and subcutaneous tissues were removed) were mounted on the Franz diffusion cells with a surface area of 0.9 cm 2 and a receptor volume of 5 mL.…”

“…HA–CH–IRN/5-FU NPs ( Figure 1A ) was fabricated by adding HA solution drop by drop into the CH–IRN/5-FU NP suspensions. 30 The obtained CH–IRN/5-FU NP and HA–CH–IRN/5-FU NP suspensions were collected by centrifugation at 10,000 rpm for 15 min, resuspended in Milli-Q water, washed three times and filtered through a 0.45 μm membrane. The obtained NPs were stored at 2°C–8°C.…”
