lncRNA GAS5 inhibits malignant progression by regulating macroautophagy and forms a negative feedback regulatory loop with the miR‑34a/mTOR/SIRT1 pathway in colorectal cancer

Abstract: Long non-coding RNA growth arrest specific 5 (GAS5) exerts inhibitory effects through the modulation of several target microRNAs (miRs) in cancer. However, its potential roles and underlying relationship during colorectal cancer (CRC) progression are unclear. Therefore, we explored the role of the negative feedback loop formed by the GAS5/miR-34a axis and mammalian target of rapamycin/sirtuin 1 (mTOR/SIRT1) pathway on macroautophagy and apoptosis in CRC. Expression of GAS5, miR-34a, SIRT1 and mTOR in CRC patie… Show more

“…Overexpression of miR-34a forms a negative feedback regulation loop through the SIRT1/mTOR/GAS5 axis, which may explain why GAS5 - mediated autophagy in CRC is in a relatively balanced state. Thus, it plays an anti-apoptotic role in cancer progression and provides a new perspective on the molecular mechanism of CRC ( 33 ).…”

“…indicated the importance of the circ_0020710/miR-370-3p/CXCL12 axis in melanoma ( 41 ). Regarding the regulated proteins, SIRT1 ( 101 103 ), CXCL12 ( 104 106 ), etc., served as important regulators in the mTOR signaling pathway.…”

“…Therefore, the use of mTOR inhibitors increases the expression of GAS5. For several cancers, the suppression of GAS5 and mTOR expression has been shown to be reciprocal [27,29]. It was suggested that these ORFs can "collect" ribosomes on themselves without protein synthesis, acting as a "sponge" for ribosomes [12].…”

“…GAS5 increases the expression of PTEN in several ways [67,68] and increases the expression of FOXO1 [54]. Moreover, in breast cancer, as in other types of cancer [27,29], the suppression of mTOR and GAS5 may be mutual. Current knowledge thus implicates GAS5 as a promising therapeutic target in breast cancer.…”
