Liver histology and histomorphometry in hybrid sorubim (Pseudoplatystoma reticulatum  × Pseudoplatystoma corruscans ) reared on intensive fish farming

Abstract: This study aimed to characterize the liver histology and histomorphometry in sorubim hybrid of different categories (nursery, growth and grow-out) reared on fish farming. The categories were defined considering body weight (BW): nursery category (n = 5): BW = 37.06 AE 6.00 g (31.6-45.3 g); growth category (n = 5): BW = 310.40 AE 53.80 g (242.1-376.4 g) and grow-out category (n = 5): BW = 874.28 AE 27.59 g (846.2-913.1 g). Liver fragments were processed to paraffin inclusion, and sections were stained by haemat… Show more

“…The liver's histomorphometry was performed by determining the number of hepatocytes per area (µm 2 ), diameter of hepatocyte nucleus (µm) and Rpcn (perimeter of the nucleus/ cytoplasm), according to Rodrigues, Saturnino, et al. (2017). Morphological alterations were evaluated qualitatively, by means of the lesion index of Bernet et al.…”

“…Intestinal villi histomorphometry was performed by measuring ten villi's height (from base to top) and width of the folds (close to apex region) per sample, and the thickness of the muscle tunic (smooth muscle) in ten points per sample, using 40x and 100x lens. Histomorphometry was performed according to Rodrigues et al (2017), from five randomly selected pictures with a 1000x magnification. The area (µm 2 ), perimeter and diameter (µm) of the hepatocytes nuclei were measured in 50 cells per fish, aiming to calculate the volume of the nucleus Vnh (µm 3 ) = 4/3 π.r 3 , where r is the nuclear ray.…”

“…Conversely, histometric techniques allow us to quantify organ structure density for a given set of histological sections (Marcos, Monteiro, & Rocha, 2012; Weibel, Kistler, & Scherle, 1966). For example, in fish, changes in hepatocyte volume are associated with metabolic status during body development, while parenchymal density may suffer seasonal and sex effects as a natural adaptative process (Rocha, Monteiro, & Pereira, 1997; Rodrigues, Saturnino, & Fernandes, 2017). In anurans, however, histometric traits have not been explored comparatively among different species.…”
