Live Imaging of the Ependymal Cilia in the Lateral Ventricles of the Mouse Brain

Abstract: Multiciliated ependymal cells line the ventricles in the adult brain. Abnormal function or structure of ependymal cilia is associated with various neurological deficits. The current ex vivo live imaging of motile ependymal cilia technique allows for a detailed study of ciliary dynamics following several steps. These steps include: mice euthanasia with carbon dioxide according to protocols of The University of Toledo's Institutional Animal Care and Use Committee (IACUC); craniectomy followed by brain removal an… Show more

“…Analysis of the localisation of adherens junctional components, including E-cadherin, β-catenin and p120, to the subapical membrane of ependymal cells would be one way in which to investigate this. Furthermore, an in-depth study of ciliary development, perhaps via electron microscopy, coupled with in vitro and ex vivo culture systems aimed at assessing ciliary function [ 1 ] would provide important insights into the functionality of ependymal cilia in Nfix −/− mice. More broadly, the use of a conditional Nfix allele [ 25 ], coupled with an ependymal-specific cre driver such as a Six3 CreER T2:GFP line will enable the role of NFIX specifically in these cells to be parsed from its role in radial glia and different mature cell types.…”

“…Brain sections from PN20-PN30 wild-type and Ttll3 −/− Ttll8 −/− mice were collected and imaged as described in (57). Briefly, the dissected brains were sectioned into ~200-mm sagittal sections in Leibovitz L15 medium.…”

“…The measurement of cilia beats was described elsewhere (Al Omran, Saternos, Liu, Nauli, & AbouAlaiwi, 2015). For high-speed imaging of ciliary beating, fresh coronal brain sections of P5 wild type or Edf1 knockout mice were placed in 30 mm glass-bottom culture dishes containing DMEM/High-Glucose supplemented with 10% fetal bovine serum (FBS) and 1% penicillin/streptomycin solution (P/S).…”
