Liposomal-based lidocaine formulation for the improvement of infiltrative buccal anaesthesia

Abstract: This study describes the encapsulation of the local anaesthetic lidocaine (LDC) in large unilamellar liposomes (LUV) prepared in a scalable procedure, with hydrogenated soybean phosphatidylcholine, cholesterol and mannitol. Structural properties of the liposomes were assessed by dynamic light scattering, nanoparticle tracking analysis and transmission electron microscopy. A modified, two-compartment Franz-cell system was used to evaluate the release kinetics of LDC from the liposomes. The in vivo anaesthetic e… Show more

“…However, after adding ABE, ABE‐liposomes became irregularly spherical and their surface turned heterogeneous and rough (Figure 1bII,bIII). Several investigators have reported the finding of liposomes with spherical shapes that are similar to our observations (de Almeida et al, 2019; Yuan et al, 2013; Yuwanda et al, 2022). In all cases, liposomes with conserved structure were observed (Battistelli et al, 2018).…”

“…All formulations were successfully prepared, showed a translucent appearance and spherical morphology (Figure 2), as previously observed with conventional liposomes of the same composition [35,36]. Encapsulation of NOR increased the size and PDI of conventional liposomes, which was not evident in PCT-containing formulations.…”

“…11c. 290 Like a previous in vivo study, liposomal articaine injected locally to infraorbital foramen caused the anesthetic affect in uninflamed tissue, but the combination of liposomal articaine and epinephrine performed better for the This journal is © The Royal Society of Chemistry 2023 inflammed tissue. 291 Similar observations have been reported for other drugs such as prilocaine, ropivacaine administered locally or applied as a gel encapsulated in lipid nanostructures at pulpal, gingival, palatal, or mucosal oral membrane in vivo or human study groups.…”
