Lipid metabolism in liver injury caused by carbon tetrachloride in the rat

Abstract: 1. A phosphoprotein (herring-egg phosvitin) has been isolated from the eggs of the herring by a procedure involving extraction with butanol-1-ol of a water-soluble lipoprotein complex.2. The calcium salt of the phosvitin contained 7 6 % of nitrogen and 12 0 % of phosphorus.3. The protein contained only five amino acids, namely serine, aspartic acid, glutamic acid, valine and threonine. Only serine was phosphorylated in the intact material.T.E.B. was a recipient of a C.N.R.S. and N.A.T.O. fellowship during this… Show more

“…In results of an early study (27) in CCl 4 -treated rats, a reduced synthesis of lipoprotein, which is responsible for transportation of triglycerides from the liver to plasma, was reported. Results of the same study suggest that accumulation of fat resulted from the destruction of mitochondrial enzymes involved in the oxidation of free fatty acids.…”

“…Hypercholesterolemia, a prominent metabolic disorder, is well recognized as a major risk factor for coronary heart disease and atherosclerosis. Although elevated serum lipid levels are often observed in liver injury (Senthilkumar et al, 2003;Aiyar et al, 1964), attention, however, has rarely been paid to its precise contribution to the pathogenesis or progression of hepatic steatosis, inflammation, and fibrosis. A few experimental studies have demonstrated an association between the intake of high cholesterol and high-fat diets with liver steatosis and inflammation (Buyssens et al, 1996;Tous et al, 2005).…”

“…In previous work from this Laboratory (Aiyar et al 1964), a decrease in the uptake of Sudan Black B by the lipoproteins of rat-liver cell sap was observed after carbon tetrachloride administration and this was interpreted as reflecting a decrease in the lipoprotein content ofthe cell sap. In the present study, in which the lipids of the cell sap were chemically determined, a considerable increase as a result of carbon tetrachloride administration was observed.…”

“…With strict adherence to this staining and washing procedure it was possible to obtain reproducible results in which the lipoproteins of the liver cell sap were fractionated into five distinct bands, instead of three as observed by Aiyar et al (1964). The diffuse staining at the origin found by Aiyar et al (1964) was avoided by the preliminary separation of the cell sap from the floating-fat fraction. All five lipoproteins were stainable with protein stains such as Bromophenol Blue and Amido Black (Jencks, Jetton & Durrum, 1955).…”
