Lipid metabolism in cancer: A systematic review

Abstract: Preclinical studies and clinical trials have emphasized the decisive role of lipid metabolism in tumor proliferation and metastasis. This systematic review aimed to explore the existing literature to evaluate the role and significance of the genes and pathways most commonly involved in the regulation of lipid metabolism in cancer. The literature search was performed as per Preferred Reporting Items for Systematic Reviews and Meta-analyses. Approximately 2396 research articles were initially selected, of which … Show more

“…However, rare research studies focus on the correlation between the prognosis of CC and FAM, which needs to be elucidated. Furthermore, the FAM can be regulated by oncogenic signalling pathway directly, namely, growth-factor receptor tyrosine kinases (RTKs)/RAS [11], phosphoinositide 3-kinase/protein kinase B (PI3K/AKT), and the mitogenactivated protein kinase (MAPK) signalling pathway [12]. However, the mechanisms driving particular FAM phenotypes in CC are still unclear.…”

“…For glycolysis, these include drugs that target hexokinase 2 (HK2), which is involved in the initial steps in glycolysis, using ATP from the mitochondria to phosphorylate glucose, or drugs that target glucose transporters (GLUT1 and GLUT4) (2,(6)(7)(8). Several drugs are also used to target lipid metabolism in cancer (9)(10)(11)(12). These include drugs that target lipid uptake (targeting proteins such as LXR, CD36 and FABP4/5), lipogenic enzymes (such as ACC, ACLY, FASN, and SCD1) and proteins involved in intracellular lipid homeostasis (such as CPT1A, PPARG and others) (13,14).…”

“…Serum lipid levels are important in many disorders' current status and progression, especially in cardiovascular and cerebrovascular diseases. In addition, recent experimental studies indicate that lipid metabolism plays a decisive role in tumor proliferation and metastases in cancer 5 . In these diseases, most of which develop on the basis of atherosclerogenic plaque, chronic inflammation is thought to be effective in plaque formation and rupture.…”

It is pointed out that in addition to classical markers such as CRP and ESR, IL-6, CXCL12 and hs-CRP can be used to define this inflammation in the laboratory.…”
