Lineage II of Southeast Asian/American DENV-2 is Associated with a Severe Dengue Outbreak in the Peruvian Amazon

Abstract: Abstract. During 2010 and 2011, the Loreto region of Peru experienced a dengue outbreak of unprecedented magnitude and severity for the region. This outbreak coincided with the reappearance of dengue virus-2 (DENV-2) in Loreto after almost 8 years. Whole-genome sequence indicated that DENV-2 from the outbreak belonged to lineage II of the southeast Asian/American genotype and was most closely related to viruses circulating in Brazil during 2007 and 2008, whereas DENV-2 previously circulating in Loreto grouped … Show more

“…Our study reports that the Panamanian DENV-2 strain, circulating during 2011 outbreak, belongs to Asian/American genotype (AS/AM). The introduction of this genotype has been linked with outbreaks with high mortality rates 41,45,46 in Peru 47 , Nicaragua 27 and Brasil 48 . Our phylogenetic analysis clustered Panama 2011 DENV-2 strains in clade 2 sub-clade 2b.I using previous classification of DENV-2 AS/AM circulating in Central America 26 , closely related with strains from Mexico 2011 and Guatemala 2009.…”

“…FFAs were conducted as described previously ( 23 ) for viral stocks, blood meals, and all mosquito samples by inoculating 96-well plates of nearly confluent Vero cells with 50 μL of sample supplemented with 50 μL of mosquito media. After a 3-day incubation, plates were fixed, washed, and blocked before overnight incubation with mouse anti– Zika virus (strain MR-766).…”

“…In Peru, a dengue outbreak during 2010 and 2011 was unprecedented in its magnitude and severity. Whole-genome sequencing documented that the responsible virus was a DENV-2 belonging to lineage II of Southeast Asian/American genotype that differed from the DENV-2 virus that had been circulating in the region since 1990 [9]. In this case, it appeared that the introduction of a new virus lineage contributed to the changed epidemiology.…”

“…These techniques have identified clades and genotypes within a serotype and determined genetic relatedness of the viruses [5,7]. Based on these phylogenetic analyses, further understanding of dengue virus origin, movement, and distribution has become possible [6,8,9].…”
