Ligustilide suppresses RANKL‐induced osteoclastogenesis and bone resorption via inhibition of RANK expression

Abstract: Osteoclast (OC) is the only cell involved in bone resorption. Dysfunction of OCs leads to a variety of bone diseases. Ligustilide (LIG) is the main component of the volatile oil isolated and purified from Angelica sinensis. LIG exerts many pharmacological activities, but its effects on osteoclastogenesis and bone resorption are still unclear. Our study showed that LIG inhibited receptor activator of nuclear factor‐κB (NF‐κB) ligand‐induced OC formation and activation in a dose‐dependent manner. Additionally, L… Show more

“…The histological results of the current study have strongly elucidated the bone regenerative capability of Angelica sinensis. This could be supported by various studies which proved that Angelica sinensis can inhibit wear debris particles-induced in ammatory bone resorption in mice [27] and can suppress osteoclastogenesis through inhibiting RANKL-mediated osteoclast differentiation in bone marrow macrophages decreasing bone resorption and preventing bone loss [28,29] .…”

“…36 In addition, the TCR-CD3 complex contains the immune receptor tyrosine-activated motif (ITAM) domain, which can be mediated with MAPK and NF-kB signals to promote the expression of c-Fos and the differentiation, maturation, and proliferation of osteoclasts, leading to bone resorption. 37,38 (Please See Figure 11)

“…Angelica sinensis has medicinal effects on depression and cognitive disorders 4 , and its active ingredients include ligustilide (LIG) 5 , ferulic acid 6 , d -glucose 7 , etc. Among them, LIG is the main component of the volatile oil of angelica sinensis 8 , which has the properties of easy volatility and easy penetration of the blood-brain barrier, which allows LIG to be delivered to the brain through intranasal administration. Therefore, LIG has a high bioavailability in the treatment of brain diseases 9 , 10 .…”
