Diazepam-based drugs are widely used today in human treatment. Diazepam may be a primary drug aimed at treating neurological diseases or an associated drug in the treatment of other diseases in the purpose of symptomatic therapy. The sedative effect of diazepam characterizes it as a drug that people usually use on their own and without a doctor’s supervision. Directly, but also through influencing the nervous system, diazepam disrupts proper functioning of all body organs. The purpose of this paper was to examine the effects of diazepam on blood and cytohistological parameters of rats in an in vivo experiment. Mallory-Azan and immunochistochemical staining methods BLX-CX and Survivin tissues of liver, kidney and spleen of rats were used to achieve the set goal. Cytometric analysis of rats detected cells in apoptosis and measurements of stereological parameters were made using a system according to Cavalier’s principle. Results of analysis of hematological and histological parameters indicate a detrimental effect of diazepam on blood parameters, as well as on structure and functioning of the liver, kidneys and spleen of rats. This paper is a foundation for further detailed scientific research with the aim of elucidating all harmful effects that diazepam has on all organs in the body of rats. This data could serve as a starting point for future studies in clinical pharmacology on therapeutic protocols for usage of diazepam-based sedatives.

To investigate the effects of diazepam, which has anticonvulsant and anxiolytic effects, and diclofenac potassium, which has anti-inflammatory, analgesic and antipyretic effects, on rat kidney tissue, used in an experimental epilepsy model. Methods: 32 Wistar albino rats (2-4 months old, 200-250 gr) were used in the study. The rats were grouped in four as 8 rats in each group: Epilepsy, Epilepsy + Diazepam, Epilepsy + Diclofenac potassium, Epilepsy + Diazepam + Diclofenac potassium. Epileptic seizure model was created with penicillin (500.000 IU) injected intracortically under urethane anesthesia. 30 minutes later, diazepam (0.1 mg/kg) and diclofenac potassium (10 mg/kg) were administered intraperitoneally. At the end of the study, rat kidneys were removed and evaluated histopathologically in terms of inflammation, glomerular shrinkage, tubular dilatation, tubular epithelial thinning, desquame epithelium, brush epithelial loss, vacuolization, hemorrhage and congestion. Results: No difference was found between diazepam and diclofenac potassium in terms of vacuolization, glomerular shrinkage, tubular dilatation and hemorrhage. Inflammation, congestion and tubular epithelial thinning rate were found to be lower inEpilepsy + Diclofenac potassium and Epilepsy + Diazepam + Diclofenac potassium group when compared with Epilepsy + Diazepam group. While brush epithelial loss and desquame epithelial rate was found to be lowest in the epilepsy group, these parameters were not found to show a significant difference between drug groups. Conclusion: It was concluded that combined use of diazepam and diclofenac potassium in their effects on kidney are more useful than their single use.

The present study was intended to find out the curative potentials of Garlic (Allium sativum) against di-(2-Ethylhexyl) phthalate (DEHP) induced toxicity in the reproductive system of female mice. Forty female mice were divided into four groups (n=10) as the (a) control group was given normal feed and drinking water, (b) aqueous garlic extract treated group (500 mg/kg), (c) DEHP group received 500 mg/kg in corn oil, and (d) DEHP + aqueous garlic extract each at a dosage of 500 mg/kg body weight. All treatments were given daily through oral gavage for 28 days. After completion of the experiment, all the animals were dissected through cervical dislocation to get reproductive organs. Collected organs were weighed and processed through the conventional histology technique of staining with eosin and hematoxylin. This study indicated that DEHP exposure caused a significant decrease (P<0.001) in body weight and weight of the complete female reproductive tract as compared to the control group, while the garlic co-administered group showed prominent improvement in body and organ weight when compared to only DEHP treatment. The adverse effect of DEHP on the histology of the ovary such as a decreased mean number of developing follicles, thin and irregular corona radiata, disruption of cumulus oophorus, and reduction in the size of the antral cavity of the tertiary follicle was observed. However, a significant recovery in the development of follicles was seen in DEHP plus garlic-treated group. DEHP plus garlic extract treatment showed protective effects on the uterus, such as a significant increase in the diameter of the uterus (P<0.001), muscularis, mean number of endometrial glands (P<0.001), and endometrial epithelial heights as compared to only DEHP exposed group. Hence, garlic extract showed significant ameliorative potential against DEHP-induced reproductive anomalies in female mice.
