Life-Sparing Effect of Human Cord Blood-Mesenchymal Stem Cells in Experimental Acute Kidney Injury

Abstract: In search for new sources of mesenchymal stem cells (MSCs) for renal repair in acute kidney injury (AKI), we investigated the potential of human cord blood (CB)-MSCs to cure mice with AKI. Infusion of CB-MSCs in immunodeficient mice with cisplatin-induced AKI ameliorated both renal function and tubular cell injury, and prolonged survival. Disclosure of potential conflicts of interest is found at the end of this article.

“…Similarly, BM-MSCs promoted PCNA expression by the tubular cells which was an index of renal regeneration as well as inhibiting cell apoptosis induced by cisplatin [31] . Furthermore, injection of human cord blood-MSCs activated the pro-survival factor Akt in the PCTs inducing their proliferation as evidenced by the high number of cells positive for PCNA staining [28] .…”

“…Thereafter, washed with phosphate buffer three times and incubated in biotinylated rabbit anti mouse IgG solution (1/400) for 20 minutes followed by streptavidin-biotin solution for 30 minutes. The antibody binding sites in the kidney sections were visualized by diaminobenzidine (DAB) and were counterstained with Mayer's hematoxylin, dehydrated, cleared and mounted [28] .…”

“…Higher therapeutic efficacy, including complete restoration of kidney function, was observed after infusion of CBMSCs/pericytes compared with regular bone marrow-derived MSCs. 62 However, few donor cells were found in the restored area; also, it was shown in culture and in vivo that the observed renoprotective effects are mediated mainly by angiogenic and antiapoptotic factors secreted by the CBMSCs/pericytes. Another promising source of stem cells is the umbilical cord itself, within which Wharton jelly (a gelatinous substance that insulates umbilical blood vessels) is a recognized source of MSCs.…”
