Levels and Determinants of Inflammatory Biomarkers in a Swiss Population-Based Sample (CoLaus Study)

Abstract: Objectiveto assess the levels and determinants of interleukin (IL)-1β, IL-6, tumour necrosis factor (TNF)-α and C-reactive protein (CRP) in a healthy Caucasian population.Methodspopulation sample of 2884 men and 3201 women aged 35 to 75. IL-1β, IL-6 and TNF-α were assessed by a multiplexed particle-based flow cytometric assay and CRP by an immunometric assay.ResultsSpearman rank correlations between duplicate cytokine measurements (N = 80) ranged between 0.89 and 0.96; intra-class correlation coefficients rang… Show more

“…In Expt A, determination of immunoreactive TNF-α in plasma and hippocampus was conducted using a flow cytometric particle based assay with a lower limit of quantification (LLOQ) of 0.5 pg/ml (Azzinnari et al, 2014;Marques-Vidal et al, 2011). For hippocampal TNF-, punches were ultrasonicated in 10x volume of complete Tris lysis buffer, centrifuged, and the supernatant pipetted off for assay.…”

“…Plasma proinflammatory cytokines tumor necrosis factor (TNF) and interleukin-6 (IL-6) were measured using a multiplexed particle-based flow cytometric cytokine assay (Marques-Vidal et al, 2011). The lower limit of detection for TNF and IL-6 was 0.5 pg/mL.…”

“…In plasma samples, the concentrations of murine TNF, IL-6 and IL-10 were determined using a multiplexed particle-based flow cytometric cytokine assay according to a published procedure (Marques-Vidal et al, 2011;Cathomas et al, 2015). For TNF, the range of the standard curve was 0.1-30 pg/ml, and the lower limit of quantification (LLOQ) was 0.3 pg/ml.…”
