Leptin receptor+ cells promote bone marrow innervation and regeneration by synthesizing nerve growth factor

Xiang Gao,

James G. Peyer
et al.

Abstract: The bone marrow contains peripheral nerves that promote haematopoietic regeneration after irradiation or chemotherapy (myeloablation), but little is known about how this is regulated. Here we found that nerve growth factor (NGF) produced by leptin receptor-expressing (LepR+) stromal cells is required to maintain nerve fibres in adult bone marrow. In nerveless bone marrow, steady-state haematopoiesis was normal but haematopoietic and vascular regeneration were impaired after myeloablation. LepR+ cells, and the … Show more

“…The absolute number of marrow cells markedly decreased after IR and remained low for up to 10 days after lethal IR in C57BL6 mice. Similarly, the frequencies of CAR/Lepr + MSCs decreased to approximately 10% of the non-IR control three days after IR, which is consistent with previous findings [25], and did not recover during our observation period (Fig. 3A and B).…”

“…4 e). Alongside vascular endothelium in the BM microenvironment, LEPR + MSCs cooperatively support HSPC function and niche integrity through the expression of hematopoietic-supportive factors [ 34 , 36 44 ]. While Bmx-creER T2 efficiently labels BM arterioles, the potential for off-target effects in LEPR + cells should be taken into consideration during experimental design and data interpretation.…”

“…BMAds and their precursors LepR + stromal cells in adult bone marrow, are the major sources of NGF, KitL and VEGF contributing to sustain nerves and promote haematopoietic and vascular regeneration after myeloablation 17 , 74 . These emerging evidences suggest marrow adipocytes homeostasis are essential, especially in impaired bone marrow environment after irradiation or chemotherapy.…”
