Abstract: Obesity is considered as a risk factor of osteoarthritis (OA), but the precise relationship is still poorly understood. Leptin, one of the most relevant factors secreted by adipose tissues, plays an important role in the pathogenesis of OA. “…Although we did not find any article about the effect of leptin on TLR4 expression in the pancreas, Jiang et al [ 51 ] observed that leptin induces TLR4 expression via the JAK2-STAT3 pathway in obesity-related osteoarthritis, so the leptin effect on TLR4 expression in other organs cannot be excluded. Moreover, Jiang et al [ 51 ] also presented that the silencing of the suppressor of cytokine signaling 3 (SOC3, the main inhibitor of leptin action) significantly influenced TLR4 expression, which can emphasize the leptin-TLR4 dependence.
