The study aimed to investigate the clinical patterns and blood profile dynamics in ewes across different reproductive stages. The research involved analyzing clinical patterns, hematological parameters, and various serum parameters from nonpregnancy to postpartum ewes, as well as mid-pregnancy ewes categorized by age (young vs. old) and litter size (single vs. twin). Analysis of serum minerals demonstrated notable changes in the levels of calcium, phosphorus, magnesium, copper, zinc, and selenium across the reproductive cycle. Calcium decreased significantly during late pregnancy (7.11±0.39mg/dL) and postpartum (9.69±0.06mg/dL), phosphorus remained consistent except for a decrease in late pregnancy (4.15±0.36mg/dL), and magnesium declined gradually from nonpregnancy to postpartum. Similarly, the examination of serum leptin, vitamin D, glucose, and triglycerides provided valuable insights into metabolic status and energy homeostasis. Leptin showed a significant increase during pregnancy (5.40±0.31, 5.80±0.06, 6.33±0.09ng/dL). The evaluation of hematological parameters revealed dynamic variations in RBC, Hb, HCT, WBC, lymphocyte, neutrophil, and MID levels throughout the reproductive stages. Additionally, the analysis of clinical patterns including body temperature, pulse rate, respiratory rate, and ruminal movement showed the adaptive changes occurring in response to pregnancy. Furthermore, comparisons between young vs. old age groups and single vs. twin lamb pregnancies indicated limited effects of age and litter size on the measured parameters. In conclusion, this research provides comprehensive insights that contribute to a better understanding of ewe health, reproductive performance, and metabolic adaptations, offering valuable information for effective management strategies, mineral supplementation, and overall care during different reproductive stages.

Comprehensive Gene Expression Profiling Analysis of Adipose Tissue in Male Individuals from Fat- and Thin-Tailed Sheep Breeds

It has been shown that tail fat content varies significantly among sheep breeds and plays a significant role in meat quality. Recently, significant efforts have been made to understand the physiological, biochemical, and genomic regulation of fat deposition in sheep tails in order to unravel the mechanisms underlying energy storage and adipose tissue lipid metabolism. RNA-seq has enabled us to provide a high-resolution snapshot of differential gene expression between fat- and thin-tailed sheep breeds. Therefore, three RNA-seq datasets were meta-analyzed for the current work to elucidate the transcriptome profile differences between them. Specifically, we identified hub genes, performed gene ontology (GO) analysis, carried out enrichment analyses of the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, and validated hub genes using machine learning algorithms. This approach revealed a total of 136 meta-genes, 39 of which were not significant in any of the individual studies, indicating the higher statistical power of the meta-analysis. Furthermore, the results derived from the use of machine learning revealed POSTN, K35, SETD4, USP29, ANKRD37, RTN2, PRG4, and LRRC4C as substantial genes that were assigned a higher weight (0.7) than other meta-genes. Among the decision tree models, the Random Forest ones surpassed the others in adipose tissue predictive power fat deposition in fat- and thin-tailed breeds (accuracy > 0.85%). In this regard, combining meta-analyses and machine learning approaches allowed for the identification of three important genes (POSTN, K35, SETD4) related to lipid metabolism, and our findings could help animal breeding strategies optimize fat-tailed breeds’ tail sizes.

Leptin is an adipokine with a pleiotropic impact on many physiological processes, including hypothalamic-pituitary-somatotropic (HPS) axis activity, which plays a key role in regulating mammalian metabolism. Leptin insensitivity/resistance is a pathological condition in humans, but in seasonal animals, it is a physiological adaptation. Therefore, these animals represent a promising model for studying this phenomenon. This study aimed to determine the influence of leptin on the activity of the HPS axis. Two in vivo experiments performed during short- and long-day photoperiods were conducted on 12 ewes per experiment, and the ewes were divided randomly into 2 groups. The arcuate nucleus, paraventricular nucleus, anterior pituitary (AP) tissues, and blood were collected. The concentration of growth hormone (GH) was measured in the blood, and the relative expression of GHRH, SST, GHRHR, SSTR1, SSTR2, SSTR3, SSTR5, LEPR, and GH was measured in the collected brain structures. The study showed that the photoperiod, and therefore leptin sensitivity, plays an important role in regulating HPS axis activity in the seasonal ewe. However, leptin influences the release of GH in a season-dependent manner, and its effect seems to be targeted at the posttranscriptional stages of GH secretion.
