Background and objectives: Recent advances in clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR associated sequences (Cas) technology has opened up immense possibilities for improving the gut health and overall immunity of the individual. In development of all these applications, lactic acid bacteria (LAB), which are already a part of human diet, are an attractive vehicle. The technology can utilize the evolutionary perspective of bacterial resistance to phages by this class of bacteria. Thus, the knowledge of CRISPR-based phage resistance in starter cultures is of interest to clinicians as well as food technologists. In the present study, an attempt has been made to explore the presence of CRISPR loci and cas gene clusters in the genomes of Lactobacilli strains available in public databases. A further analysis has been undertaken to identify the spacers left behind by the bacteriophages encountered by Lactobacilli during their evolution. Materials and methods: A total of 174 completed and draft genomes of Lactobacilli strains were analyzed by different online tools like CRISPR-Cas finder and CRISPR-Cas++. Results: Different types of the CRISPR-Cas system found in 58 genomes out of 174 genomes were analyzed. No CRISPR sequences were found in 109 genomes. The analysis yielded type I and type II CRISPR-Cas system in 14 genomes each and type III in 1 genome. The study found 32 bacteriophage spacers in different bacterial genomes that predict the identity of phages infecting the bacterium over its evolutionary history. Interpretation and conclusion:This study is an exploratory one that has predicted the presence of CRISPRs and their diversity across Lactobacillus species.

Abstract-Bacteriocins are antimicrobial peptides that help bacteria fight competing bacteria in microecological systems. Bacteriocins of lactic acid bacteria (LAB) have attracted much interest in recent years because of their properties that make them suitable as natural food preservatives against specific food pathogens, and as possible supplement to antibiotics against drug resistant bacterial strains. LAB bacteriocins are generally classified into the lantibiotics and non-lantibiotics, the latter divided into four subgroups. To date, only nisin and to a lesser extent, pediocin are the commercially applied bacteriocins for food use. Clinical applications are still limited to animal health. One of the more exciting prospects on the use of bacteriocins is the possibility of subjecting them to bioengineering to either increase antimicrobial activity or further specify their target microorganism. The latter would make it less damaging to the natural gut microflora, which is a common drawback of conventional antibiotic therapy. This paper focuses on the nature, biology, and applications of bacteriocins based on knowledge gained abroad and in the Philippines during the last two decades.

The consumption of probiotic microorganisms in fermented products has been used for centuries. Lactobacillus spp. is one of the main species studied due to its various beneficial effects on health. This species has a great ability to adapt to hostile environments, produces antimicrobial substances capable of destroying or inhibiting the growth of pathogenic bacteria, and is involved in the digestion of complex carbohydrates not digested by the host. However, there are still some uncertainties and disagreements about the precise biochemical metabolism of lactobacilli. The aim of this article is to review the metabolism and physiological characteristics of lactobacilli for a better understanding of the benefits that these bacteria promote in the host and for the development of strains and probiotic products with higher health benefits.
