Lactate Dehydrogenase B Controls Lysosome Activity and Autophagy in Cancer

Abstract: Metabolic adaptability is essential for tumor progression and includes cooperation between cancer cells with different metabolic phenotypes. Optimal glucose supply to glycolytic cancer cells occurs when oxidative cancer cells use lactate preferentially to glucose. However, using lactate instead of glucose mimics glucose deprivation, and glucose starvation induces autophagy. We report that lactate sustains autophagy in cancer. In cancer cells preferentially to normal cells, lactate dehydrogenase B (LDHB), catal… Show more

“…Thus, exposure to PPAR-γ ligands following the inflammatory response may be the mechanism by which the inflammation is resolved. 6 It is intriguing that lactate (pK a 3.86) supports lysosomal acidification and autophagy in tumour cells [172].…”

“…Actually, the basal autophagy activity is essential for cell homeostasis, involving cell growth, survival, development and death. The levels of autophagy must be precisely regulated, as indicated by the fact that dysfunctional autophagy has been related to wide ranges of human diseases, such as cancer [24] [25], neurodegenerative diseases [26], myopathies [27], gastrointestinal disorders [28], heart and liver diseases [29]. The most interesting is the study about autophagy and cancer.…”
