Ohtake et al (1994) reported that seed storage-protein composition was altered in nonnodulating soybean mutants, and that little or no accumulation of the b-subunit of b-conglycinin could be observed in the mutants. As they were unable to ®x nitrogen, nitrogen supply to the mutant plants was greatly reduced and the relative sulfur supply was therefore higher than in the wild-type plants.…”

“…Free amino acids were extracted three times with 80% ethanol and the contents were determined using the ninhydrin method (Ohyama 1990), while the contents of free sugars were determined using the phenol-sulfuric acid method (Osaki 1990). Protein extraction and sodium dodecylsulfate-polyacrylamide gel electrophoresis were carried out as previously described (Ohtake et al 1994).
