Laboratory and clinical evaluation of conjugate vaccines composed of Staphylococcus aureus type 5 and type 8 capsular polysaccharides bound to Pseudomonas aeruginosa recombinant exoprotein A

Abstract: The synthesis, standardization, and immunogenicity in young outbred mice and clinical evaluation in adult volunteers of investigational vaccines designed to induce serum antibodies to the type 5 and type 8 capsular polysaccharides (CPs) of Staphylococcus aureus are described. Conjugates composed of the type 5 CP and a sonicated preparation of a high-molecular-weight type 8 CP bound to a nontoxic recombinant protein derived from Pseudomonas aeruginosa exotoxin A (rEPA) were synthesized. The conjugates were nont… Show more

“…Capsular serotyping of the coded isolates (no patient identifiers were used) was performed by direct cell agglutination and immunoprecipitation of cell extracts as described previously (21). Two different types of rabbit serum samples were used to identify the capsular type of each isolate: sera from animals vaccinated with a whole-cell vaccine (21) and monospecific sera against either type 5 or type 8 capsular polysaccharide conjugated vaccine prepared in rabbits (11,12). Monoclonal antibodies to capsular types 5 and 8 produced and standardized by NABI (Rockville, Md.)…”

“…In the experiments reported here, two proteins of medical interest with lower molecular weights than TT, the B subunit of the heat-labile cholera-like enterotoxin (LT-B) of Escherichia coli and the recombinant exoprotein A (rEPA) of Pseudomonas aeruginosa, were used as carriers for Vi (7,8,14,15). The immunogenicity of these conjugates in young mice and guinea pigs and their safety in adult volunteers are described.…”

“…In the mouse model, antibodies elicited by the S. aureus bivalent T5/ 8 conjugate vaccine are said to be functional and a high degree of correlation has been reported to exist between antibody titer by ELISA and opsonic activity by in vitro opsonophagocytosis. 8 The T5 and T8 PS-specific antibodies from both vaccinated humans and animals have been shown to induce type-specific opsonophagocytic killing of S. aureus, 29,30 and are protective in both lethality and sublethality mouse models. 31 Thus serum IgG responses to T5/ 8 PS by ELISA have been used as surrogate markers for the protectiveness of this vaccine.…”
