Abstract:The aim of this work was to formulate O/W/O multiple emulsions containing açaí oil as a model system and to evaluate their physical stability and in vivo Sun Protection Factor (SPF). Multiple emulsions are complex dispersion systems, known also as, "emulsions of emulsions". These emulsion systems, have significant potential in the cosmetic industry. Euterpe oleracea Mart., Arecaceae, popularly known in Brazil as "açaí", is an economically important plant. Açaí oil has been used as antioxidant and as anti-inflammatory activities. The multiple emulsions were prepared using a two-step procedure. The investigated formulations were characterized and their stability over time was evaluated by preliminary and accelerated stability. O/W/O multiple emulsions containing the same concentration of sunscreens with and without açaí oil were evaluated by the International Sun Protection Factor Test Method. The samples containing 70% (w/w) of primary emulsion, 5% (w/w) PEG-30-dipolyhydroxystearate, 10% (w/w) of açaí oil and 5% (w/w) of sucrose polybehenate have been found to be stable. The rheological measurements revealed that the samples exhibited non-Newtonian pseudoplastic flow behavior and thixotropy. To conclude, no statistical difference could be observed on the in vivo SPF to both multiple systems with or without açaí oil.

Caracterizadas por serem um sistema carreador complexo contendo duas emulsões distintas que coexistem simultaneamente, as emulsões múltiplas apresentam muitas vantagens e aplicações, contudo manter a sua estabilidade apresenta-se ainda como um grande desafio. Foi realizada uma revisão da literatura existente além de uma análise sobre os principais fatores que interferem no controle da estabilidade da emulsão múltipla. Dentre os fatores destaca-se a presença de eletrólitos, a concentração do componente ativo osmótico, a influência dos aditivos estabilizantes, a natureza da fase oleosa, a propriedade dos filmes interfaciais e a relação de volume entre as fases, assim como o método de preparo. A observação desses fatores pode propiciar a elaboração e o uso das emulsões múltiplas de maneira proveitosa e propiciar a sua maior aplicabilidade em produtos farmacêuticos.

The aim of the present study was to develop water-in-oil-in-water (w/o/w) type Multiple Emulsion of Lisinopril dihydrate for enhancing its oral bioavailability via enhanced permeation. For primary emulsification, corn oil was used as the oil phase, Span 83 as the lipophilic surfactant and Xanthan gum was used as the viscosity enhancer. Primary emulsion was re-emulsified with aqueous phase containing Tween 20 as hydrophilic surfactant. Preliminary screening was performed using a 12-run, 8-factor, 2-level Plackett-Burman design followed by Box Behnken Design for optimization. MEs were characterized and evaluated for macroscopic and microscopic properties, globule size, entrapment efficiency, rheological properties, in vitro and ex vivo drug release and stability studies. In vitro drug diffusion study was done through dialysis bag and ex vivo permeability studies were performed in Franz diffusion cell using rat intestine. The optimized w/o/w ME showed globule size and entrapment efficiency of 15.65 + 1.967 µm and 87.35 + 3.79 % respectively. Drug flux was found to be 119.3 µg/cm2/h for drug loaded w/o/w ME and 105.1µg/cm2/h for plain drug solution. The overall results of the studies showed the potential of the w/o/w ME as promising drug delivery system for Lisinopril dihydrate.
