Kidney Tissue Targeted Metabolic Profiling of Unilateral Ureteral Obstruction Rats by NMR

Abstract: Renal interstitial fibrosis is a common pathological process in the progression of kidney disease. A nuclear magnetic resonance (NMR) based metabolomic approach was used to analyze the kidney tissues of rats with renal interstitial fibrosis (RIF), induced by unilateral ureteral obstruction (UUO). The combination of a variety of statistical methods were used to screen out 14 significantly changed potential metabolites, which are related with multiple biochemical processes including amino acid metabolism, adenin… Show more

“…The typical 1 H - NMR spectra of cell and medium are present in Supplementary Figure S6 . Resonance assignments (Supplementary Table S1 ) were performed based on the chemical shifts of standard compounds from the Chenomx NMR suite, the Human Metabolome Database (HMDB) 18 , and Biological Magnetic Resonance Data Bank (BMRB) 19 , as well as the literature data 20 , 21 . The NMR spectra of cell and medium were dominated by peaks from amino acids, organic acids, choline-containing metabolites, and amine metabolites, in addition to some other metabolites.…”

“…Representative 600 MHz 1 H NMR spectra of plasma and tissue extracts obtained from control and HD on 15th days were showed in Figure 2 , with major metabolites in the integrated regions assigned. NMR spectral data was analyzed and specific metabolites were identified by the previously reported reference (Kim et al, 2014 ; Guo et al, 2015 ; Liao et al, 2016 ; Li et al, 2016 ) and in-house NMR database and the database of Chenomx NMR Suite 6.0 (Chenomx, Edmonton, Canada). Metabolites detected in plasma, tissue extracts including amino acids, glycolysis products, tricarboxylic acid cycle (TCA cycle) intermediates and choline metabolites, etc.…”

“…31 Studies have shown that unilateral ureteral obstruction in rats signicantly decreases the valine levels in the body by enhancing the protein synthesis activities to repair damaged membrane protein structures. 32 We determined a signicant reduction in the serum valine at the end of the 15 th and 23 rd days possibly due to the destruction of cell membrane proteins by hydrocortisone. Under normal circumstances, through the alanine-glucose cycle of toxic ammonia in the muscle, a non-toxic form of alanine may be formed and transported to the liver, where urea is synthesized from two ammonia molecules and one carbon dioxide molecule before excretion.…”
