KDM6A promotes chondrogenic differentiation of periodontal ligament stem cells by demethylation of SOX9

Abstract: KDM6A is required in chondrogenic differentiation of PDLSCs by demethylation of H3K27me3, and EZH2 inhibitor could rescue chondrogenesis of PDLSCs after knockdown of KDM6A. It could be inferred that upregulation of KDM6A or application of EZH2 inhibitor might improve mesenchymal stem cell mediated cartilage regeneration in inflammatory tissue destruction such as osteoarthritis.

“…miR‐204 was reported to function as an inhibitory molecule in oral squamous cell carcinoma by suppressing invasiveness and cancer stemness of tumours via targeting Slug, epithelial‐mesenchymal transition of tumours via Sox4 and initiation or progression of cancers via C‐X‐C motif chemokine receptor 4 . In addition, miR‐204 may also be involved in the chondrogenic differentiation of periodontal ligament stem cells . miR‐513c was reported to inhibit the development of nervous system tumours and hepatocellular carcinoma .…”

“…Osteoarthritis (OA), which is characterized by osteophyte formation, slight synovial inflammation, subchondral bone lesions, and destruction of articular cartilage, is the most common chronic joint disease. [1][2][3] The incidence of OA has been demonstrated to increase with age, especially in patients over the age of 65 years. [4][5][6] It is estimated that more than 100 million people are affected by OA worldwide.…”

“…In addition to affecting osteogenesis and adipogenesis, the H3K27me3 demethylases KDM6A and KDM6B have also been reported to facilitate the chondrogenic differentiation of MSCs [84][85][86][87]. A study on Kdm6b −/− mice showed that KDM6B is recruited to the promoter of Runx2, a critical regulator of chondrocyte maturation and endochondral bone formation, and Ihh and activates the expression of these genes by decreasing the levels of repressive H3K27me3 marks [85].…”
