iTRAQ-based quantitative proteomics analysis of cantaloupe (Cucumis melo var. saccharinus) after cold storage

Abstract: Background: Cantaloupe is susceptible to cold stress when it is stored at low temperatures, resulting in the loss of edible and commercial quality. To ascertain the molecular mechanisms of low temperatures resistance in cantaloupe, a cold-sensitive cultivar, Golden Empress-308 (GE) and a cold-tolerant cultivar, Jia Shi-310 (JS), were selected in parallel for iTRAQ quantitative proteomic analysis. Results: The two kinds of commercial cultivars were exposed to a temperature of 0.5°C for 0, 12 and 24 days. We fou… Show more

“…Protein data analysis was performed according to previous description ( Song et al, 2020 ). Briefly, the raw data were converted into mgf files via Proteome Discoverer 1.4 (Thermo, American), the mgf data were further analyzed with Protein Pilot 5.0 (AB Sciex, United States).…”

“…Gas chromatography/mass spectrometry (GC/MS) was optimized for amino acids as previously described ( Song et al, 2020 ). Raw data analysis was finished with Chroma TOF (V 4.3x,LECO) software, and the LECO-Fiehn Rtx5 database was utilized for metabolite identification by matching the mass spectrum and retention index ( Shah et al, 2020 ).…”

“…Based on our previous study, we find that GSTs are positively expressed in cold-tolerant Hami melon (JS) and are regarded as candidate cold-resistance proteins ( Song et al, 2020 ). Herein, a comprehensive study was carried out to identify the Hami melon GST gene family on the aspect of bioinformatics analysis, including molecular information, distribution on chromosomes, phylogenetic relationships, synteny relationships, gene structures, protein–protein interactions (PPIs), 3-D models, functional annotation and expression levels of total enzyme activities, and genes in response to cold stress.…”

“…Our results showed that glutathione S-transferase F11 (GSTF11) was upregulated in both OG and TE, while glutathione S-transferase (DHAR3) and (DHAR2) were upregulated in OG only. Glutathione S-transferase DHAR3 and glutathione S-transferase DHAR2-like proteins were upregulated in cantaloupe crops in response to cold stress [ 65 ]. Nonetheless, as antioxidant agents, peroxiredoxin (Prx) and thioredoxin play a vital role in redox signaling [ 61 , 66 ].…”
