Duck plague (DP) is the most feared duck disease in the world. For isolation, identification, molecular detection and characterization of DP virus (DPV), a total of 94 samples were collected from commercial farms (n=6) and households (n=13) from Rajshahi (n=37), Netrokona (n=35) and Mymensingh (n=22) districts of Bangladesh. The samples were processed and inoculated into 11-13 days old embryonated duck eggs for virus propagation. Virus was identified using agar gel immunodiffusion test (AGIT) and passive hemagglutination (PHA) test, and was confirmed by polymerase chain reaction (PCR) targeting DNA polymerase and gC genes, followed by sequencing. Pathogenicity tests were performed using duck embryos, ducklings and ducks. Among the 94 samples, 17 isolates were confirmed as DPV by PCR amplification of partial DNA polymerase (446bp) and gC genes (78-bp), respectively. One of the isolates (Anatid herpes 1 BAU DMH) was sequenced and found to be closely related with a Chinese variant of DPV (GenBank: JQ647509.1). Thus, we assume that both Bangladeshi and Chinese isolates of DPV may have a common ancestor.

The present study was carried out for the isolation and molecular detection of duck plague virus (DPV) for the development of inactivated vaccine seed from the local outbreaks. A total of 12 suspected dead duck samples were collected from commercial farms and local market at Sunamganj, Netrokona and Mymensingh districts. Then, the samples were processed and prepared inocula were inoculated into 9-12 days old duck embryonated eggs. In duck embryonated eggs, several passages (3-4) were performed before infection into DEF cell culture. Presence of viral DNA was confirmed by PCR using the primer for DNA polymerase gene. After PCR confirmation, virus cultured in DEF cell was used for the preparation of formalin (0.12%) inactivated and oil based adjuvanted vaccine and was experimentally injected to 18 ducklings and 5 were kept as control. TCID50 of the selected virus for vaccine preparation was 108.70/ml. The mean passive haemagglutination assay (PHA) titre of sera of samples at 0 days, 7 days and 14 days post vaccination were 4.0±0, 14.22±1.78 and 44.44±4.4, respectively, which indicated significant (p<0.01) increase of antibody titre. Embryonated duck eggs and DEF cell culture are effective for virus isolation and on the basis of PHA test, it could also be suggested that the experimentally developed DP vaccine can be used successfully for the prevention of DP in Bangladesh. Duck viral enteritis (DVE) is an acute and contagious herpes virus infection with a potential threat to domestic and wild waterfowl. In this study, 75% mortality rate with 40% drop in egg production was recorded among the examined flocks. Diagnosis of the disease was carried out based on the clinical signs, pathology and supported by laboratory confirmation. Microscopically, the presence of eosinophilic intranuclear inclusion bodies (IN/IB) in the hepatocytes revealed duck enteritis virus (DEV) infection. DEV was isolated in embryonated duck eggs; 42.6% of the samples showed evidence of infection. The nucleic acid of the DEV was detected by Polymerase Chain Reaction (PCR) in 19 out of 68 collected samples (27.9%) with positive amplification of the DNA polymerase gene. This investigation disclosed variation in the disease prevalence among the different ages, breeds, seasons, and immune status of ducks, as the disease was more prevalent among vaccinated flocks (34.8%) than in non-vaccinated ones (24.4%). Accordingly, control efforts can be done to reduce the further outbreaks of DVE in terms of adoption of good management practice, biosecurity measures and increasing awareness about the disease spread and application of proper vaccination regimens. Continuous studies of DEV at the molecular and immunological levels are important for the effectiveness of any control strategy of the disease including vaccination.Key words: DVE; duck plaque; histopathology; PCR; DNA polymerase gene; Egypt
