ABSTRACT.Western blotting was performed to analyze Neospora caninum tachyzoite antigens recognized by mouse IgG at different stages of infection including recrudescence. At the early stage of infection, a 36-38 kDa antigen was clearly recognized by the mouse antisera. After day 48 postinoculation, the signal of the 36-38 kDa antigen gradually weakened. Meanwhile, a 43 kDa antigen was intensely and continuously recognized from 48 to 125 days postinoculation. This 43 kDa antigen was clearly detectable with the antisera from the mice under immunosuppression. Sera from naturally infected cattle strongly reacted with the 43 kDa antigen. Therefore, the 43 kDa antigen may be useful for immunological reactions to detect infected animals except in the early stage of the infection.

The impact of in vitro isolation and molecular characterisation of Neospora caninum as well as sequence analyses was studied. The brain homogenate of a naturally Neospora-infected dairy cow (positive in ELISA and Western blot) was intraperitoneally inoculated into Mongolian gerbils (Meriones unguiculatus). The brain of gerbils on day 60 post-inoculation was homogenized, and, after trypsin-digestion, cultured on Vero cells. Neospora-like tachyzoites were first observed after 77 days of cultivation. The parasite was confirmed by polymerase chain reaction (PCR) using Neospora-specific primers Np21 and Np6. The PCR product of the first Slovak isolate (NC-SKB1) was subsequently sequenced and published in GenBank under accession number GU300774. Sequencing and BLAST search identified the isolate as N. caninum.

ResumoA neosporose, doença emergente causada pelo Neospora caninum, é considerada como a principal causa de abortamentos em bovinos em vários países. Estudos têm sido realizados até hoje com diversas espécies de animais, visando melhor elucidação da neosporose considerando agente, hospedeiro e meio ambiente. Esta revisão enfatiza aspectos sobre patogenia e diagnóstico da neosporose em alguns animais domésticos. Palavras-chave: Neospora caninum. Bovinos. Cães. Ovinos. Eqüídeos. AbstractNeosporosis, emerging disease caused by Neospora caninum was considered the most important causes of aborts in cattle from many countries. Some studies have been developed in several animal species to understand host-parasite relationship. The present review describes some aspects about pathogeny and diagnostic of the neosporosis in domestic animals.
