Isolation of Mononuclear Cells From Tonsillar Tissue

Abstract: This unit describes a protocol for the derivation of lymphoid cell populations from tonsillar tissue. The procedure can also be applied with little or no modification to spleen and lymph node tissue. Human tonsils are the most readily available lymphoid organs and are often used as a source of large numbers of cells characteristic of local lymphoid tissue. Mononuclear cells are isolated by mechanical disruption of tonsil tissue followed by Ficoll-Hypaque gradient centrifugation. Cells may then be separated int… Show more

“…Human PBMCs were isolated from buffy coats by density gradient centrifugation using Ficoll‐Paque PLUS (GE Healthcare). Mononuclear cells (MCs) from human tonsils were isolated from patients undergoing tonsillectomy as previously described . After density gradient centrifugation using Ficoll‐Paque PLUS, ILCs were enriched by using magnetic cell depletion of CD3 + T cells with CD3 microbeads and LD columns (Miltenyi) according to the manufacturer's instructions.…”

“…Written informed consent was obtained from all patients prior to sample acquisition and experiments have been approved by the Ethics Committee of the Charité Medical University, Berlin (EA2‐078‐16, EA1/149/1). Mononuclear cells (MCs) from human tonsils were isolated from patients undergoing tonsillectomy as previously described . After mashing and density gradient centrifugation using Ficoll‐Paque PLUS, ILCs were enriched by using magnetic cell depletion of CD3 + T cells with CD3 mAb microbeads and LD columns (Miltenyi) according to the manufacturer's instructions.…”

“…Cell populations from human tonsils were isolated from patients undergoing tonsillectomy, as previously described (Ryon, 2001). Gut LP cells were isolated from CRC or IBD patients undergoing colectomy, by using a modified version of the protocol of Bull and Bookman (Bull and Bookman, 1977).…”
