The safety of raw milk sold in Northern Italy was investigated in relation to hygiene quality parameters and presence of Salmonella spp., Listeria monocytogenes, thermotolerant Campylobacter, and Verocytotoxin producing Escherichia coli O157:H7. The performance of different analytical methods used-official culture method (ISO), modified Bacteriological Analytical Manual cultural method (mBAM), and polymerase chain reaction (PCR)-was evaluated. The presence of Mycobacterium avium subsp. paratuberculosis (Map) was investigated only by PCR. All samples met regulations for alkaline phosphatase and inhibitory substance, while 18% and 44.8% of samples collected from vending machines had, respectively, somatic cell count (SCC) > 300,000/mL and total bacterial count (TBC) > 50,000 CFU/mL. The correlation between hygienic quality parameters in samples collected from bulk tank and vending machines showed a significant increase of TBC in vending machines meaning that raw milk was mishandled during distribution and sale. All pathogens investigated were detected in raw milk sold at vending machines; a total of five samples (5%) had at least one pathogen, of which two were detected by PCR and three by mBAM. None of the samples was positive by cultural ISO methods. Even if the comparison of analytical methods showed that none performs significantly better than the others, testing a higher volume of milk (25 versus 210 mL) affects significantly the detection rate of pathogens. Three samples (3%) were positive for Map, suggesting that raw milk is a significant source of Map exposure for consumers. The observed TBC increase and the detection of several pathogenic bacteria pose questions on the safety of raw milk; the use of ISO seems inefficient in detecting a low contamination level of pathogens in milk and consequently not appropriate as official method for testing. In order to ensure consumer's safety, a new approach for the raw milk chain is required.

IntroductionEscherichia coli O157 and other enterohemorrhagic E. coli (EHEC) are a worldwide threat to public health (1). Mostly in developed countries, E. coli O157 has been recognized as a cause of serious clinical symptoms such as hemorrhagic colitis (HC) and hemolytic uremic syndrome (HUS) characterized by thrombocytopenia, hemolytic anemia, and nephropathy (2). Cattle are the primary reservoir and asymptomatic carriers of E. coli O157. The bacteria were spread via cattle faces (3). Most outbreaks of EHEC infections have been associated with the consumption of uncooked meat and meat products, unpasteurized dairy products, and feces-contaminated vegetables and water (2-4).E. coli O157, a member of EHEC, has important virulence factors such as shiga toxins (stx1 and stx2), enterohemolysin (hlyA), and intimin (eaeA). Shiga toxinproducing E. coli (STEC) can cause HC and HUS (5,6) due to the inhibiting protein synthesis, eukaryotic cell injury, and death (7). Intimin, encoded by the eaeA gene, is able to adhere to intestinal mucosa and cause intestinal lesions (5,7). In addition, enterohemolysin, encoded by hlyA, leads to lysis of the erythrocytes, which may contribute to the iron intake of the bacterium to live in the intestine (4).The frequency of antimicrobial resistance in foodborne pathogens such as E. coli, Salmonella spp., and Campylobacter spp. has recently increased. The misuse of antimicrobial agents for farming and therapeutic purpose in animals and humans is the main cause of transmission of antibiotic-resistant strains, which are very difficult to treat with commonly used antibiotics, to humans via the food supply (8).The aim of this study was to investigate the potential risk of public health by determining the prevalence of E. coli O157:H7 and identifying the virulence genes from The aims of this study were to investigate the prevalence of E. coli O157 and to identify virulence genes and also to determine the antibiotic susceptibility of the isolates in different poultry-originated materials.

Escherichia coli produtoras de toxina de Shiga (STEC) são patógenos veiculados por alimentos capazes de causar desde diarréia branda até severa e sanguinolenta e evoluir para complicações graves como colite hemorrágica, síndrome hemolítica urêmica e púrpura trombótica trombocitopênica. Esses microrganismos têm sido associados a numerosos surtos e vários casos esporádicos de infecções em todo o mundo devido ao consumo de alimentos contaminados. O sorogrupo O157 desse grupo de microrganismos é considerado o principal devido ao seu envolvimento em surtos de doença por STEC, entretanto, muitos casos vêm ocorrendo em todo o mundo devido a cepas patogênicas de STEC não-O157, como O26, O103, O111 e O145. Os objetivos do presente trabalho foram avaliar a ocorrência de STEC em três pontos da linha de abate de bovinos destinados à exportação e em cortes refrigerados de aves e de bovinos comercializados na região da Grande São Paulo; pesquisar a presença dos fatores de virulência dos isolados através dos genes stx 1 , stx 2 , eaeA e ehxA; identificar isolados de E. coli O157:H7 pela pesquisa dos genes uidA, rfb O157 e flic H7 ; verificar a citotoxicidade em células Vero; pesquisar a atividade enterohemolítica dos isolados; avaliar o perfil de suscetibilidade a antimicrobianos; identificar os sorotipos e avaliar a diversidade genética dos isolados de STEC obtidos. Na linha de abate, 201 animais foram amostrados, obtendo-se 603 amostras que compreenderam 201 amostras provenientes do couro, 201 de carcaça e 201 de meia carcaça. No varejo, foram analisadas 100 amostras de cortes de carne bovina e 100 de cortes de frango. A metodologia utilizada para detecção de E. coli sorogrupo O157 foi a preconizada pela ISO 16654, enquanto para os sorogrupos O26, O103, O111 e O145 foi empregada a metodologia descrita pelo "Surveillance Group for Diseases and Infections of Animals" (NRM 006). Os isolados obtidos foram confirmados como STEC pela técnica de PCR. Dos 201 animais amostrados, dois (1,0%) foram positivos para STEC, obtendo-se sete isolados (três do animal número 399 e quatro do animal 401) do couro. Não houve o isolamento do microrganismo nas amostras de carcaça e meia carcaça. Os sete isolados apresentaram o perfil stx 2 , uidA, eaeA, ehxA, rfb O157 e fliC H7 podendo, assim, ser considerados E. coli enterohemorrágica (EHEC) pertencentes ao sorotipo O157:H7.
