-This study estimated the genetic diversity and structure of 12 genetic groups (GG) of locally adapted and specialized pigs in the state of Pernambuco using 22 microsatellite markers. Nine locally adapted breeds (Baé, Caruncho, Canastra, Canastrão, Mamelado, Moura, Nilo, Piau and UDB (Undefined Breed)) and 3 specialized breeds (Duroc, Landrace and Large White), totaling 190 animals, were analyzed. The Analysis of Molecular Variance (AMOVA) showed that 3.2% of the total variation was due to differences between genetic groups, and 3.6% to differences between local and commercial pigs. One hundred and ninety eight alleles were identified and apart from the Large White breed, all GG presented HardyWeinberg Equilibrium deviations for some loci. The total and effective allele means were lower for Duroc (3.65 and 3.01) and higher for UDB (8.89 and 4.53) and Canastra (8.61 and 4.58). Using Nei's standard genetic distance and the UPGMA method, it was possible to observe that the Landrace breed was grouped with the local genetic groups Canastra, Moura, Canastrão, Baé and Caruncho. Due to the complex admixture pattern, the genetic variability of the 12 genetic groups can be analyzed by distributing the individuals into two populations as demonstrated by a Bayesian analysis, corroborating the results from AMOVA, which revealed a low level of genetic differentiation between the inferred populations.

________________________________________________________________________________ AbstractThis paper details genetic characterization and trends from a microsatellite-based study of genetic diversity on southern African pig populations. A total of 351 pigs from three commercial breeds and three indigenous populations were genotyped at 39 loci. Differences among the levels of genetic diversity in populations correlated well with known population histories. In commercial breeds, heterozygosity was higher in the well established SA Landrace and Large White breeds (0.580 and 0.636) compared to the Duroc breed, established more recently (0.531). In indigenous populations, the highest heterozygosity levels were found in the Mozambican and South African populations (0.692 and 0.634) with a lower value of 0.531 in a smaller Namibian population. A hierarchical division of total genetic diversity revealed a high betweenpopulation component of 17.9%. F ST -and R ST -based analysis confirmed high levels of differentiation, with pair-wise comparisons between breeds indicating significant differentiation in 20 out of 21 comparisons.Results from an assignment test confirmed results from F ST and R ST and suggested a true genetic structure with significant differentiation between most populations sampled, but with little differentiation among the commercial SA Landrace and Large White breeds. The results are discussed with reference to known historical information on commercial and indigenous pig populations. This paper also presents new data on the optimization of microsatellite markers for application in Sus scrofa domestica.

Wild pigs (Sus scrofa) are the most widely distributed invasive wild ungulate in the United States, yet the factors that influence wild pig dispersal and colonization at the regional level are poorly understood. Our objective was to use a population genetic approach to describe patterns of dispersal and colonization among populations to gain a greater understanding of the invasion process contributing to the expansion of this species. We used 52 microsatellite loci to produce individual genotypes for 482 swine sampled at 39 locations between 2014 and 2016. Our data revealed the existence of genetically distinct subpopulations (F ST = 0.1170, p \ 0.05). We found evidence of both fine-scale subdivision among the sampling locations, as well as evidence of long term genetic isolation. Several locations exhibited significant admixture (interbreeding) suggesting frequent mixing of individuals among locations; up to 14% of animals were immigrants from other populations. This pattern of admixture suggested successive rounds of human-assisted translocation and subsequent expansion across Florida.
