Isolation, identification and characterization of Burkholderia pseudomallei from soil of coastal region of India

Abstract: Melioidosis is an emerging infectious disease caused by a free living soil dwelling Gram-negative bacterium Burkholderia pseudomallei. The disease is endemic to most parts of Southeast Asia and northern Australia and the organism has been isolated from moist soil and water. In India clinical cases are recently reported from the states of Tamilnadu, Kerala, Karnataka, Maharashtra, Orissa, Assam, West Bengal, Pondicherry and Tripura. This study is aimed to confirm the prevalence of this important bacterial speci… Show more

“…Similarly to our findings of B. pseudomallei isolation from mangrove, it has been commonly reported the microorganism isolation from moist soil and water (Prakash et al, 2014;Knappik et al, 2015;Liu et al, 2015). Also, the high pluviometric index in the Amapá state (Segovia et al, 2011), as part of the Amazon region of Brazil, contributes to maintain the elevated soil humidity and natural collections of water.…”

“…PCR amplification was performed with primer set FC27 (5′-AGAGTTTGATCCTGGCTCAG-3′) and RC1492 (5′-TACGGCTACCTTACGACTT-3′) (Sigma Aldrich, USA). The methodology for sequencing was adapted from the earlier reports [ 21 ]. BLAST search was performed using NCBI database and multiple sequence alignment was carried out in ClustalX [ 22 ].…”

“…21 In a recent publication, it was been isolated from soil in the eastern coastal part of India. 9 In an unpublished study, B. pseudomallei has also been isolated from soil in Mangalore. The intensity of rainfall has shown to be an independent risk factor for pneumonia, septic shock and death.…”

“…3 Recently, B. pseudomallei was isolated from soil in the eastern coastal region of India, thereby defining the geographical area in which humans and animals have an increased risk of melioidosis infection. 9 Studies have shown a positive correlation between the incidence of melioidosis and monsoon rains in endemic areas. 1 Environment studies have also found increased melioidosis cases among paddy workers.…”
