The wide spectra of colonizing microorganism, likewise Burkholderia pseudomallei, has been found in different habitats, and presenting distinct activity, as exerting physiological functions among plants, or as a pathogen for man, animals and also as a phytopathogen. A common disease of men and animals caused by B. pseudomallei, melioidosis, is a severe morbidity that usually culminates in the host death. Soil samples from different areas in the Amapá state, in northern Brazil, were screened for environmental microorganisms to assess potential antimicrobial activity aiming at biotechnological applications. Among the prospected microorganisms, B. pseudomallei was isolated from high humidity soils, mangrove, which is rich in organic materials, produced by the diversified local flora and fauna. The isolated B. pseudomallei was identified by its biochemical profile and growth characteristics. Molecular confirmation of B. pseudomallei phenotypic identification was achieved by PCR amplification of the 16 S ribosomal DNA. The sequencing of amplified products confirmed that the Amapá sample, and two other isolates from human infections in Ceará state, northeast Brazil, were B. pseudomallei, and sequence alignement to the same specie, MSHR146 strain from Australia, and clone YN01 from uncultured Burkholderia sp., deposited in the GenBank, exhibited close phylogenetic relationship among them. Until now, there is no report of B. pseudomallei related disease among human and animal populations in the Amapá state, despite the finding of B. pseudomallei in it, in an area of water buffalo ranching and flowing small rivers utilized by human populations.

The polycyclic aromatic hydrocarbons (PAHs) pollution to the environment is a major threat to the living organisms, and hence the degradation of these PAHs is necessary. Studies on PAHs degrading bacteria have focussed on terrestrial microbes and the potential of marine derived microbes is undermined. Herein we report the isolation and characterization of PAHs degrading Burkholderia sp. from lagoon sediments collected at the Southern coast of India. The strain was Gram negative, rod-shaped, motile, and ∼2–5 μm in length. Based on the phylogenetic data the strain was identified as Burkholderia and designated as VITRSB1. Initial PAHs degradation ability of the strain was assessed using basal salt medium supplemented with diesel, kerosene, toluene, aniline, naphthalene, and phenol. The strain was found to be effectively degrading kerosene, diesel, toluene, and aniline even at higher concentration (1%). However, naphthalene and aniline were degraded only at lower concentration (0.1%) and phenol, camphor, and DAP inhibited the growth of the strain. Furthermore, the degraded end products of the PAHs were determined using FTIR. Notably, none of the end products were found to be toxic to the biosphere. Our results indicate that the isolated Burkholderia sp. could be a prospective candidate for the effective degradation of selective PAHs.

Objective: To determine the geographical epidemiology, clinical presentations and risk factors associated with melioidosis. Methods: A total of 28 confirmed cultures of Burkholderia pseudomallei were isolated and prospectively analysed with respect to clinico-demographic factors. Results: Age groups ranged from neonate to 84 years of age. Five children were less than 12 years of age. There was a male predominance of 71.4%. There was a crude mortality of 25%; all deaths were due to septic shock, 42.8% of which occurred within 48 hours of admission. A strong linear association (p = 0.01) was found between intensity of rainfall and presentation of cases. Proportion of cases and mortality were greater among those with occupational or recreational exposure to soil (75%). Pneumonia (32.1%) was the most frequent primary clinical presentation and diabetes mellitus (64.3%) constituted a major risk factor for both development and death due to melioidosis. Higher occurrence of mortality was noted among patients with chronic kidney disease (CKD) than others (Fischer's exact test p = 0.04). One case of recurrence was noted. Conclusion: Melioidosis is quite prevalent in the western coastal region of India and is strongly associated with intensity of rainfall. There is increased risk among diabetics and to those who are exposed to soil and surface water. Melioidotic lymphadenopathy may mimic tuberculosis, hence should be considered as a differential diagnosis. Patients with bacteraemia have a poorer prognosis; and, septic shock relates to imminent death.
