Isolation, Culture and Identification of Choriocarcinoma Stem-Like Cells from the Human Choriocarcinoma Cell-Line JEG-3

Abstract: Background/Aims: Cancer stem cells (CSCs) exhibit enhanced proliferative capacity and resistance to chemotherapy; however, choriocarcinoma CSCs have not yet been reported. In this study the human choriocarcinoma cell line JEG-3 was cultured in serum free media, and the characteristics of suspension and parental adherent JEG-3 cells were compared. Methods: Cell proliferation, colony-formation, soft agar clonogenicity, and transwell invasion assays were performed in vitro, and tumor xenografts in BALB/c nude mic… Show more

“…Hoechst staining is toxic to cells, and CSCs isolated via the SP cell sorting method may exhibit decreased self-renewal, tumorigenicity and other CSC properties; thus, the use of this technique is limited (36). FACS and MACS require knowledge of specific surface markers of CSCs, and questions have been raised regarding the specificity and reliability of markers for the identification of CSCs as widely accepted specific surface markers of CSCs have not yet been defined (37). Binding of an antibody to its receptor during these two methods may result in cell activation, influencing the biology of cells (38).…”

“…Based on preliminary experiments set up to optimize the kinetics of JEG-3 spheroid growth, JEG-3 cells at 80% confluence were seeded at 3 × 10 4 cells/ml in complete RPMI medium additioned with 1.5% agarose [19], allowing to obtain spheroids of 100-250 μm in diameter at 2-4 days of culture. JEG-3 spheroid size was measured using an optic microscope equipped with a calibrated eyepiece reticule.…”

“…CSC-like cells from the human choriocarcinoma cell line JEG-3 were isolated and identified in this study [21]. The study aimed to examine the effect of functional AhR on the development of CSC-like cells, identify their characteristics, and verify the molecular mechanisms.…”
