Isolation and Screening of Filamentous Fungi Producing Extracellular Lipase with Potential in Biodiesel Production

Abstract: Nineteen fungal strains were isolated from a chicken slaughterhouse effluent and within those, only one showed high values of lipolytic activity in submerged cultures. This fungus was identified as Trametes hirsuta. The crude extract was immobilized in chitosan/clay beads, with an immobilization yield of 80.9%. The analyses of the crude extract and the immobilized derivative at different temperatures, pH (s), solvents, metallic ions and storage showed that the immobilization process increased the enzyme life s… Show more

“…However, our findings could not compare to the hydrolysis activity of filamentous fungi isolates reported by Pacheco et al. () who reported activities ranging from 89 U/ml to 361 U/ml, using various substrates. Lipase production was targeted in this study because lipase based technologies for the synthesis of novel compounds is on the increase (Andualema & Gessesse, ).…”

“…In this regard, Sharma et al (2001) reported that trace elements assist with structural components and are stabilizers of enzymes, whereas ions such as Mg 2+ , Fe 2+ , Ca 2+ , Cu 2+ , Co 2+ , Na + , K + , Mn 2+ , and Zn 2+ appear to positively influence lipase production. The effect of the salts on the properties of the proteins is perceived by the activity, conformational stability, and solubility [44]. These effects may appear due to ionic bonds at specific sites of the proteins, altering their level of hydration [45].…”

“…Other micronutrients needed to regulate these functions are potassium, considered essential for osmoregulation [47], iron, which in turn is essentially used for synthesis of heme and cytochrome [48], and calcium, considered necessary for the effective stabilization of lipases [49]. These characteristics can be associated with structural changes of the enzyme and the fact that the salts increase the dispersion of enzymatic molecules, facilitating the mass transfer during the reaction [44].…”

“…A blank sample was always used containing medium described in Maia et al [ 16 ] instead of enzyme solution. The molar extinction coefficient of ε = 15,000 M −1 cm −1 for pNP at 410 nm [ 17 , 18 ] was used for calculus. One enzymatic unit was defined as the amount of the enzyme that releases one mmol of pNP in one minute under the assay conditions.…”
