Isolation and Molecular Identification of Leishmania spp. Agents in Patients with Cutaneous Leishmaniasis in Yazd Province, Endemic Region of Central Iran

Abstract: Background: Cutaneous Leishmaniasis (CL) is a major health problem in many parts of Iran. Many methods have been introduced for detection and identification of Cutaneous Leishmaniasis. The purpose of this study was isolation and molecular identification of Leishmania spp. agents in patients with CL from endemic region of central Iran. In this study, one of the main loci of central Iran named Yazd will be assessed CL identification using PCR-RFLP. Methods: For this cross-sectional study, sampling was done from … Show more

“…35 Eslami et al in Yazd, Iran, identified 49.01% of samples to be L. major and 50.98% to be L. tropica. 36 People traveling and moving over the time have caused epidemiological changes and created new foci in some parts of Iran. For example, a focus of L. major was created in Pakdasht, Tehran due to people traveling to Sabzevar city (a focus of L. major).…”

“…Twice a week, sandflies from inside buildings (bedrooms, covered warehouses, and corridors) and the opened areas (rodent nests, cracks in walls, and bird nests) were caught using 30 glue traps (white papers measuring 15 × 20 cm impregnated with the castor oil). 23 In all cases, the next morning after trapping, adhesive traps were collected before sunrise and transferred to the laboratory for the identification of the mosquitoes species. For this purpose, first, they were identified by entomological experts using organological tests, and then the sandflies were carefully collected from the traps with a dissection needle.…”

“…To detect and identify Leishmania spp., a diagnostic key based on restriction banding pattern of ITS1 was used based on specific primer pair of LITSr-F 5′-CTG GAT CAT TTT CCgATg-3′ and L5.8 s-R 5′-TGA TAC CAC TTA TCG CAC TT-3′ [20]. Amplification reaction mixture included 1x PCR buffer, 0.2 mM dNTP, 1.5 mM MgCl 2 , 1.5 U Taq DNA polymerase, 0.5 µM each primer pair, and 100 ng DNA.…”
