Isolation and molecular characterization of type I and type II feline coronavirus in Malaysia

Abstract: BackgroundFeline infectious peritonitis virus (FIPV) and feline enteric coronavirus (FECV) are two important coronaviruses of domestic cat worldwide. Although FCoV is prevalent among cats; the fastidious nature of type I FCoV to grow on cell culture has limited further studies on tissue tropism and pathogenesis of FCoV. While several studies reported serological evidence for FCoV in Malaysia, neither the circulating FCoV isolated nor its biotypes determined. This study for the first time, describes the isolati… Show more

“…Type I FCoV is the dominant genotype in most countries, including Austria, Malaysia and China [19, 22, 32], while type II is less common and is mainly found in southeast Asian countries. Co-infection with types I and II were reported sporadically in Europe a decade ago and in southeast Asia in recent years but has not been reported in mainland China [20, 23, 25, 32].…”

“…FCoV has a worldwide distribution [19–25]. Infections with FCoV are mostly type I FCoV, while type II FCoV occurs primarily in Southeast Asia [20, 22, 23].…”

“…Three-hundred ml of body effusion samples and suspensions of rectal swab samples from FIP-suspected cases were prepared as 10% (w/v) suspensions with sterile phosphate-buffered saline. The fluid was clarified by centrifugation at 1,000 g for 10 min, and the supernatant was collected [ 27 ]. Viral RNA was extracted using a viral RNA purification kit according to the manufacturer’s instructions (E.Z.N.A.…”

“…The main distinction between them is associated with their differences in the S gene sequence and antibody neutralization properties ( Hohdatsu et al, 1991 ; Jaimes et al, 2020 ; Shiba et al, 2007 ; Tekes and Thiel, 2016 ). Serotype FCoV-I is 80-90% more prevalent than FCoV-II worldwide ( Amer et al, 2012 ; Kummrow et al, 2005 ; Myrrha et al, 2019 ). The emergence of serotype FCoV-II has involved recombination events between the S and M genes of FCoV-I and CCoV-II ( Herrewegh et al, 1998 ; Terada et al, 2014 ).…”
