Cellulose is the basic component of lignocellulosic biomass (LCB) making it a suitable substrate for bioethanol fermentation. Cellulolytic and ethanologenic bacteria possess cellulases that convert cellulose to glucose, which in turn yields ethanol subsequently. Heterotermes indicola is a subterranean termite that causes destructive damage by consuming wooden structures of infrastructure, LCB products, etc. Prospectively, the study envisioned the screening of cellulolytic and ethanologenic bacteria from the termite gut. Twenty six bacterial strains (H1− H26) based on varied colonial morphologies were isolated. Bacterial cellulolytic activity was tested biochemically. Marked gas production in the form of bubbles (0.1−4 cm) in Durham tubes was observed in H3, H7, H13, H15, H17, H21, and H22. Sugar degradation of all isolates was indicated by pink to maroon color development with the tetrazolium salt. Hallow zones (0.42−11 mm) by Congo red staining was exhibited by all strains except H2, H7, H8, and H19. Among the 26 bacterial isolates, 12 strains were identified as efficient cellulolytic bacteria. CMCase activity and ethanol titer of all isolates varied from 1.30 ± 0.03 (H13) to 1.83 ± 0.01 (H21) umol/mL/min and 2.36 ± 0.01 (H25) to 7.00 ± 0.01 (H21) g/L, respectively. Likewise, isolate H21 exhibited an ethanol yield of 0.40 ± 0.10 g/g with 78.38 ± 2.05% fermentation efficiency. Molecular characterization of four strains, Staphylococcus sp. H13, Acinetobacter baumanni H17, Acinetobacter sp. H21, and Acinetobacter nosocomialis H22, were based on the maximum cellulolytic index and the ethanol yield. H. indicola harbor promising and novel bacteria with a natural cellulolytic tendency for efficient bioconversion of LCB to value-added products. Hence, the selected cellulolytic bacteria can become an excellent addition for use in enzyme purification, composting, and production of biofuel at large.

The Sugar Factory of Tjoekir Jombang is one of the sugar factories located in Jombang, East Java under PT Perkebunan Nusantara X. Sugarcane waste, also known as bagasse causes pollution of the environment. Some microorganisms are able to degrade cellulose-containing bagasse, because of cellulolytic enzymes produced by cellulolytic bacteria. This study aims to obtain cellulolytic bacteria isolates, screen the highest cellulolytic activity, identify the highest cellulolytic activity isolates, and optimize conditions (pH and temperature) for the highest cellulase activity. Cellulolytic bacteria from bagasse were grown on a medium containing 1% CMC. Several additional minerals were other than those in the CMC medium. The cellulase activity was assayed semi-quantitatively with the addition of 0.1% Congo red and quantitatively using the 3,5-Dinitro Salicylic Acid (DNS) method. Bacterial isolates with high cellulolytic activity were identified based on the 16S rDNA sequence. This research obtained 20 bacterial isolates, where isolate A1T4 had the highest cellulolytic index of 1.18 mm. Measurement of cellulase activity using the DNS method showed that isolate A2T2 had the highest cellulase activity of 2.19 U/mL. Hemolysis assay showed that from 12 isolates, only two isolates have γ-hemolysis activity (isolates A1T6 and A3T3). Those isolates were optimized in a CMC broth medium with temperatures of 30°C and 37°C and pH 5 and pH 6 to produce the highest cellulase activity. Isolate A1T6 and A3T3 were grown optimally at 30oC and pH 6. Isolate A1T6 was identified as Citrobacter amalonaticus with a similarity of 99.80%, and isolate A3T3 was identified as Pseudomonas mendocina with a similarity of 98.83%.

The development of renewable energy resources is strongly urged to recoup the shortage of fossil-based energy and its associated pollution issues. Energy production from carbohydrate materials has recently been of great interest due to the availability, reliability, and abundance of carbohydrate sources. Significantly, the catalytic transformation of waste carbohydrates into furanbased biofuels, specifically 2,5-dimethylfuran (DMF), appears to be an attractive solution to the aforementioned energy and environmental issues. The potential of DMF as a renewable fuel is prospective, with its physicochemical properties that are similar to those of fossil fuels. Therefore, the current work focuses on the production of DMF, with the important aspects for enhanced DMF yield being summarized herein. Notably, the significant catalysts derived from zeolite, noble-metal, non-noble-metal, metal− organic framework, and electrocatalytic materials are discussed, alongside their effects in deriving carbohydrates to DMF. Furthermore, the mechanisms of DMF production were clarified too, followed by the scrutinization of the effects from reaction conditions, solvents, and hydrogen donors onto the DMF yield. Finally, the purification process, commercialization potential, and economic feasibility of DMF production were incorporated too, with insightful future directions being identified at the end of our review. This review is expected to advocate DMF production from carbohydrate materials, which could alleviate the energy and environmental problems encountered presently.
