Biocatalytic potential of Streptomyces strains isolated from the rhizosphere of plants and from mycorrhizosphere of fungi has been investigated. A total of 118 Streptomyces isolates were selected and functionally screened for 10 different biotechnologically important enzymatic activities: hydrolase (cellulase, cutinase, gelatinase, lipase, protease, polyhydroxyalkanoate (PHA) depolymerase), phenol oxidase and peroxidase (laccase, tyrosinase, and lignin peroxidase), and aminotransferase. Out of 118 tested Streptomyces spp., 90% showed at least one enzymatic activity. The most abundant were enzymes involved in the biomass degradation, as the production of cutinase, cellulase, and lignin peroxidase were detected in 31%, 40%, and 48% of the isolates, respectively. The improved specific activities of lipase (isolates BV315 and BV100) and tyrosinase (isolates BV87 and BV88) were shown in comparison with the industrially relevant activities of Pseudomonas strains. Plant rhizosphere soils were more prolific source of Streptomyces strains with biocatalytic potential in comparison with mycorrhizosphere soils. Overall, 284 enzyme activities among 118 Streptomyces isolates have been detected. This is the first comprehensive screening of Streptomyces isolates from rhizosphere and mycorrhizosphere soils for novel biocatalysts, showing that specific environmental habitats, such as rhizosphere soils, are "treasure troves" of Streptomyces with biocatalytic potential.

Lipases are hydrolytic enzymes responsible for the hydrolysis of triacylglycerol into glycerol and free fatty acids. The biodiversity of crude oil contaminated soil was explored for the isolation of novel potent lipase producing microorganisms. In this current study, microorganisms isolated from crude oil contaminated soils were screened for lipase activity and expression. Six fungal strains namely: Yeast, Aspergillus flavus, Aspergillus niger, Verticillus sp., Penicillum sp., and Microsporum audouini demonstrated lipase producing potentials and the best two: Verticillus sp. and Penicillum sp. were selected for enzyme production and characterization. The lipase enzyme was produced in broth medium. Optimization of selected biochemical parameters reveal that optimum temperature for lipase activity was 40 ºC, pH 7 with an incubation time of 168 h for lipase expressed by Verticillus sp. and 40 ºC, pH 8 with an incubation time of 144 h was established for lipase expressed by Penicillium sp. The kinetic parameters of the lipase enzyme revealed that the enzymes had low Km (0.19 and 0.30 respectively) for the selected substrate.

Tyrosinase is a copper-containing metalloprotein that catalyzes the oxidation of tyrosine, in particular, L-DOPA to L-Dopaquinone, which are precursors of brown pigments in some wounded eukaryotic tissues. The present study focused on screening, production and characterization of tyrosinase from multi-enzyme producing Fusarium solani B1 and Fumago sp. A total of 25 strains were isolated from rotting wood samples and screened for hydrolytic and oxidative multi-enzyme potentials using different polymeric substrates. The two most consistent strains: Fusarium solani B1 and Fumago sp. B13 were further evaluated for tyrosinase production. Some media cultural parameters and physiological conditions were optimized in order to maximize tyrosinase production. Incubation of Fumago sp. B13 and Fusarium solani B1 for 96 and 144 h in medium containing 2 % and 0.2 % ratios of Glucose and NaNO3 with pH 6 and 7, respectively, was most suitable for tyrosinase production. Characterization of the partially purified tyrosinase from Fumago sp. B13 and Fusarium solani B1 exhibited optimal activities at pH 6-7, 30 °C, and 1 mM Cu2+, respectively, thereby suggesting their potentials for novel biotechnological applications.
