Isolation and Characterization of Lipoprotein Profiles in Newborns by Density Gradient Ultracentrifugation

Abstract: Summarvborn infants were selected and neonates with history of chronic Lipoproteins in newborn plasma were isolated from a minimal sample amount (0.3 ml) by a single-step ultracentrifugation in a density gradient, spanning the density range 1.02-1.20 g/ml. After 66 h ultracentrifugation in a swinging-bucket rotor, the content of the tube was eluted and collected in 0.4 ml fractions. Cholesterol and apoproteins AI, AII, and B were assayed in each fraction yielding both the distribution and composition of the ve… Show more

“…The concentration of this protein in cord blood is approximately one-half to two-thirds that of adults (25)(26)(27)(28). In adult plasma, the (HDL,,),, region is characterized by the presence of a large proportion of particles containing apo A1 without apo A11 (29).…”

“…Lipoproteins were isolated by isopycnic ultracentrifugation from 0.3 ml plasma (8). As apo E is released from lipoproteins during ultracentrifugation, 0.5 ml was also fractionated by gel filtration on a Sepharose 6B-CL column, enabling the separation of triglyceride-rich lipoproteins, LDL, and HDL (9).…”

“…On the basis of animal experiments, lipoprotein lipase activity of newborn infants has been suggested to be low at birth and to increase postnatally (1,6,7). If so, infants at birth might be incapable of using fat effectively for energy.…”
